SFRP1/FZD6抑制Wnt/β-catenin信号通路对A549肺癌细胞生物学行为的影响

摘　　要：目的研究分泌型frizzled相关蛋白1(secreted frizzled-related protein 1,SFRP1)影响A549肺癌细胞Wnt/β-catenin信号通路的机制及其对细胞增殖、克隆形成和迁移侵袭能力的影响。方法 q RT-PCR和western blot检测SFRP1在肺腺癌及癌旁组织中的表达。在A549细胞中通过质粒过表达SFRP1和用si RNA敲低FZD6的表达,采用q RT-PCR、western blot检测Wnt信号通路相关基因的表达水平变化。通过CCK-8、平板克隆形成、划痕实验以及transwell实验分别检测SFRP1/FZD6对A549细胞增殖、克隆形成、细胞迁移侵袭能力的影响。结果 SFRP1在肺癌组织中表达水平显著低于癌旁组织(P<0.05);过表达SFRP1显著抑制了A549肺癌细胞中Wnt/β-catenin信号通路,干扰受体FZD6的表达能够显著减弱SFRP1对Wnt/β-catenin信号通路的抑制效应。过表达SFRP1显著抑制肺癌细胞的增殖、克隆形成和迁移侵袭能力等恶性表型,并且能够通过干扰FZD6被逆转。结论 SFRP1通过受体FZD6抑制A549肺癌细胞的Wnt/β-catenin信号通路活化,进而抑制细胞的增殖、克隆形成和迁移侵袭能力。SFRP1/FZD6可能在肺癌进展中发挥重要作用,靶向SFRP1/FZD6有望成为研发治疗肺癌的重要靶点。Objective to study mechanism of SFRP1/FZD6 influencing signaling pathway of Wnt/beta-catenin of A549 lung cancer cells and its effect on cell proliferation,colony formation and migration and invasion ability. Methods detect expression of SFRP1 in lung adenocarcinoma and adjacent tissues by q RT-PCR and Western blot. For A549 cells,overexpress SFRP1 through plasmid,and lower expression of FZD6 with si RNA,detect expression level changes of Wnt signaling pathway related genes by q RTPCR,western blot,detect effect of SFRP1/FZD6 on proliferation,clone formation and cell migration and invasion ability of A549 cells respectively by CCK-8,plate colony formation,scratch test and transwell test. Results expression level of SFRP1 in lung cancer tissues was significantly lower than adjacent tissues（P〈0.05）; overexpressed SFRP1 significantly inhibited Wnt/beta-catenin signaling pathway of A549 lung cancer cells,expression of FZD6 receptor can significantly weaken inhibitive effect of SFRP1 for Wnt/beta-catenin signaling pathway. Overexpressed SFRP1 can significantly inhibit malignant phenotypes of proliferation of lung cancer cells,clone formation and migration invasion ability,and can be reversed by interfering FZD6. Conclusion through FZD6 receptor,SFRP1 can inhibit activation of Wnt/beta-catenin signaling pathway of A549 lung cancer cells,and then inhibit proliferation of cells,clone formation,migration and invasion ability. SFRP1/FZD6 may play an important role in progress of lung cancer,targeted SFRP1/FZD6 may be an important target of research and development of lung cancer.
