摘 要:本研究旨为从兔粪中分离、筛选出酶活性较高的产纤维素酶菌,并对该细菌进行形态学的初步鉴定。利用分离培养基(LB)从普通家兔新鲜粪便样品中分离出细菌群,经纯化后接种到产纤维素酶菌筛选培养基中。经一段时间培养后用0.1%刚果红染色液对筛选培养基进行染色,用1 mol/L氯化钠溶液进行脱色处理,根据培养基中水解透明圈直径与菌圈直径的比值挑选出酶活性较强的产纤维素酶菌株。然后,利用革兰氏染色法进行初步鉴定,利用光学显微镜观察其革兰氏染色的结果及其细菌形态。结果表明:从兔粪中分离并筛选出了具有较高酶活性的产纤维素酶菌菌株SK1和SK6,鉴定后认为是革兰氏阳性菌(G+菌),属于杆菌。研究提示,菌株SK1和SK6的水解效果显著,具有较高的开发价值与应用前景。The purpose of this study was to isolate cellulose producing bacteria with higher enzyme activity from rabbit manure, and to further identify morphology of this bacteria. In the present study, the bacterial groups were isolated from rabbit manure using isolation medium (LB), and inoculated to culture medium for cellulase producing strain after purification. Following a period of culture, the selection mediums were stained with 0.1% Congo red coloration and decolorized with 1 mol/L NaCI solution. Strain groups with higher enzyme activity were selected according to the diameter ratio of hydrolysis transparent circle to bacteria circle. Furthermore, the strain was primarily identified by Gram's stain, and the results of stain and bacterial morphology were observed using light microscope. The results showed that positive bacteria (G+bacteria) SK1 and SK6 with higher enzyme activity belonging to BaciUus were successfully isolated and screened from rabbit manure. SK1 and SK6 showed significant hydrolysis effects, and had high development value and application prospect.
