摘 要:研究虎杖苷对低氧性肺动脉高压的作用,探讨虎杖苷对防治肺动脉高压的作用机制。SD雄性大鼠实验分为三组:正常组,低氧组,低氧+虎杖苷组。通过病理染色观察肺组织改变,体外培肺动脉养平滑肌细胞,MTT检测细胞活力,Western blot检测增殖蛋白的表达。病理染色发现低氧肺血管中膜增厚,血管重构;MTT分析显示虎杖苷能显著降低缺氧PASMCs细胞活性。Western blot检测结果显示,虎杖苷抑制细胞增殖蛋白PCNA的表达。虎杖苷能抑制平滑肌细胞增殖,改善缺氧肺动脉血管的重构,可能开发作为肺动脉高压的治疗药物。Effects of polydatin on hypoxic pulmonary hypertension were investigated and the mechanism on prevention and treatment of pulmonary hypertension by polydatin was explored.Rats were randomly divided into 3 groups: control group,hypoxia group,and hypoxia+polydatin(PD) group.Pulmonary artery smooth muscle cells(PASMCs) were isolated by enzymatic method and culture.Cell viability was analyzed by MTr and the proliferation protein was detected using Western blot.Pathological examination revealed that the hypoxia group had hypertrophy of vascular media and pulmonary vascular remodeling.MTr analysis showed that PD could signifi- cantly decrease the viability of hypoxic PASMCs.Western blot showed that PD inhibited the expression of proliferating cell nuclear antigen(PCNA).These results suggested that PD could inhibit the preliferation of smooth muscle cells and improve the remodeling of hypoxic pulmonary arteries, therefore it might be developed as a therapeutic agent for pulmonary hypertension.
