青光安Ⅱ号方对慢性高眼压大鼠视网膜热休克蛋白的影响

摘　　要：目的通过建立慢性高眼压(EIOP)大鼠模型,观察青光安Ⅱ号方对慢性EIOP大鼠视网膜热休克蛋白(HSP)的影响。方法将40只SD大鼠随机抽取6只作为空白组,其余均采用烧灼巩膜表浅静脉法建立大鼠慢性高眼压模型,监测造模后大鼠的眼压,使其维持在25 mm Hg(1 mm Hg=0.133 k Pa)以上持续8周视为造模成功。将造模成功的大鼠按随机数字表法分为5组,每组6只,分别为:模型组、青光安Ⅱ号方低、中、高剂量组、益脉康组,并开始灌胃。灌胃后14 d、28 d分别处死大鼠,光镜下观察各组视网膜形态学改变,Western blot检测HSP27、HSP60、HSP70的表达。结果 Western blot:三种HSP在正常视网膜中均有表达;在EIOP模型中,三种HSP在视网膜中的表达均上升,差异有统计学意义(P<0.05)。灌胃后14 d:青光安Ⅱ号方低、中、高剂量组、益脉康组大鼠的视网膜HSP27、HSP6及HSP70的表达均增多,与模型组比较,差异均有统计学意义(P<0.05),其中,HSP27的表达,青光安Ⅱ号方高剂量组效果优于益脉康组,差异有统计学意义(P<0.05);HSP60及HSP70的表达各灌药组比较,差异无统计学意义(P>0.05)。灌胃后28 d:各灌药组HSP表达量与模型组比较,差异有统计学意义(P<0.05);其中,青光安Ⅱ号方高剂量组效果优于益脉康组,差异有统计学意义(P<0.05)。结论 (1)烧灼巩膜表浅静脉法建立大鼠EIOP模型成功率较高,眼压能有效的维持在高眼压状态;(2)EIOP大鼠视网膜中HSP27,HSP60,HSP70的表达均会应激性增多;(3)与模型组比较,青光安Ⅱ号方能促进实验性EIOP大鼠HSP27、HSP60、HSP70的表达;(4)通过实验观察初步说明青光安Ⅱ号方高剂量组对慢性高眼压大鼠视神经的保护作用优于益脉康分散片。OBJECTIVE To observe the effect of Qingguang＇an Ⅱ formula on heat shock protein（HSP） of the chronic elevated intraocular pressure（EIOP） cases through rat model. METHODS Firstly, 6 out of 40 male SD rats were randomly selected and allocated to the blank group（group A）. Then, the remained 34 male SD rats were used to establish the chronic EIOP model by cauterizing superficial scleral vein. Rats with intraocular pressure above25 mm Hg and lasted for more than 8 weeks were deemed as qualified models. After that, the 34 modelrats were divided into 5 groups evenly by random number table as model group（group B）, low dose of Qingguang＇an Ⅱ group（group C）, medium dose of Qingguang＇an Ⅱ group（group D）, high dose of Qingguangan Ⅱ group（group E）, Yimaikang group（group F）. After that, the rats were treated by intragastric administration.These animals were sacrificed 14 or 28 days after intervention for specimen of retina to study the morphological changes by light microscope and expression of HSP27, HSP60 and HSP70 by western blot. RESULTS Western blot result showed all 3 kinds of HSP expressed in normal retina.After model establishment, the expression of the 3 kinds of HSP rose sharply and the difference was statistically significant（P〈0.05）. Fourteen days after intragastric administration,the expressions of HSP27, HSP60 and HSP70 in group C, D, E, F were higher than it of group B,the difference was statistically significant（P〈0.05）. The expression of HSP27 of group E was higher than it of group F, the difference was statistically significant（P〈0.05）.Comparison of HSP60 and HSP70 among intervention groups showed no significant difference（P〈0.05）. Twenty-eight days after intragastric administration, in contrast to model group, express of 3 HSP in intervention group indicated statistically difference（P〈0.05）.
