开导加补益针刺治疗对形觉剥夺大鼠F-VEP及神经元的影响  被引量:2

摘  要:目的观察以"开导之后宜补论"为理论基础的针刺治疗方法对形觉剥夺大鼠闪光视觉诱发电位(F-VEP)及神经元的影响,并探讨其可能的作用机制。方法出生后14 d的健康Wistar大鼠36只,随机分为6组,除正常对照组外均予单眼睑缝合术制作单眼形觉剥夺弱视动物模型。造模动物于术后第7天开始治疗,阳性对照组予左旋多巴(20 mg/kg)灌胃;开导针刺组予开导针刺法,针刺太阳穴、上星穴;补益针刺组予补益针刺法,针刺脾俞穴、肾俞穴,并攒竹穴透刺睛明穴;开导加补益针刺组先进行开导针刺,接着进行补益针刺治疗。干预治疗7 d后对实验动物进行F-VEP检查,采用尼氏染色法对大鼠视皮层神经元进行形态学观察。结果 F-VEP:阴性对照组P_2波振幅较正常对照组明显降低(P<0.05),潜伏期较正常对照组明显延长(P<0.05),阳性对照组、开导针刺组、补益针刺组及开导加补益针刺组的P_2波振幅、潜伏期均明显好于阴性对照组,其中开导加补益针刺组的波形指标最好,其P_2波振幅及潜伏期好于阳性对照组(P<0.05)并与正常对照组接近(P>0.05)。视皮层组织切片:阴性对照组视皮层神经元数量较正常组明显减少(P<0.05),阳性对照组视皮层神经元数量较阴性对照组明显增加(P<0.05),与正常对照组接近(P>0.05);开导针刺组和补益针刺组视皮层神经元数量较阴性对照组明显增加(P<0.05),但低于阳性对照组(P<0.05);而开导加补益针刺组视皮层神经元数量明显多于阴性对照组(P<0.05),与正常对照组和阳性治疗组无明显差别(P>0.05)。结论开导加补益针刺治疗对单眼形觉剥夺性弱视大鼠F-VEP波形及神经元损伤具有明显的改善作用。OBJECTIVE To observe effects of acupuncture treatment based on tonifying after dredging method for form deprivation rats model in terms of F-VEP and neuron and further to discuss its possible mechanism.METHODS Thirty-six Wistar rats were randomly divided into 6 groups(six in each group) by suturing the eyelid of one eye 14 days after birth. The rats in all groups were treated 7 days after modeling. Specifically, rats in positive control group were treated with oral administration of L-dopa at the dose of 20 mg/kg. The rats in dredging acupuncture group were needled in Taiyang and Shangxing(GV23). Rats in tonifying acupuncture group were treated with acupuncture in Shenshu(BL23), Pishu(BL20), Zanzhu(BL2) through Qingming(BL1) acupoints. Flash visual evoked potential(F-VEP) method and Nissl staining was used to observe effects of the dredging acupuncture, tonifying acupuncture, dredging plus tonifying acupuncture on function of visual cortex in monocular form deprivation ambly-opia rats and morphology of visual cortex. RESULTS Dredging acupuncture, tonifying acupuncture and dredging plus tonifying acupuncture improved the F-VEP wave form and alleviated neuron damage in monocular form-deprived amblyopia rats. The result of Nissl staining indicated that the dredging plus tonifying acupuncture improved the morphology of the visual cortex in amblyopia eyes with monocular form deprivation. CONCLUSIONS Acupuncture in dredging plus tonifying method improved the F-VEP waveform and alleviated neuron damage in monocular form-deprived amblyopia rats.
