摘 要:目的:探讨喜炎平注射液对大鼠体内外及人血体外抗血小板聚集作用及其对凝血功能、凝血因子活性的影响。方法:以二磷酸腺苷(ADP)作为诱导剂,测定喜炎平注射液在大鼠体内外及健康人血体外给药对血小板5min之内的最大聚集率;并测定喜炎平注射液对健康人体外全血凝血象和凝血因子活性水平。结果:喜炎平注射液在SD大鼠体内给药剂量400、800mg/kg,体外血药浓度在3.13mg/ml、6.25mg/ml时血小板聚集率与空白对照组比较,可显著性降低血小板5min之内最大聚集率,抑制率与给药剂量呈正相关;健康人体外血药浓度在2.5mg/ml下血小板5min之内最大聚集率与对照组比较,可显著性降低5min之内血小板最大聚集率,抑制率明显提高。健康人的凝血酶原时间(PT)、活化部分凝血活酶时间(APTT)、凝血酶时间(TT)与自身空白对照组比较,时间均有不同程度的延长,具有显著性差异。纤维蛋白(Fi B-C)含量减低、抗凝血酶III(AT-III)活性明显下降。纤维蛋白降解产物(FDP)含量显著性增多。凝血因子II、V、VII、VIII、IX、X、XI、XII的活性与自身空白组比较,均有不同程度的显著降低。结论:喜炎平注射液具有抗血小板聚集作用和抗凝血作用,同时又通过降低AT-III活性来促进凝血的作用,因此,喜炎平注射液对凝血系统既有抗凝也有促凝的双重效果。Objective: To investigate influences of Xiyanping on the platelet aggregation,coagulation and coagulation factor in SD rats in vivo and human. Methods: Adenosine diphosphate(ADP) was the inducer. The platelet maximum aggregation rate and the maximum inhibition rate were determined after Xiyanping injection was administered in SD rats in vivo and in vitro human blood was administered for 5 min; Levels of the coagulation,fibrinolysis and coagulation factor activity were detected. Results: 400 mg/kg and 800 mg/kg Xiyanping,compared with the control group(P〈0. 05,P〈0. 01),significantly reduced the 5 min maximum aggregation rate,the inhibition rate was positively correlated with the dosage; when in vitro blood concentrations of rats were at 3. 13 mg/ml and 6. 25 mg/ml,the 5 min platelet aggregation rate compared with the control group(P〈0. 01),significantly decreased,the inhibition rate was positively correlated with the dosage; when the blood concentration at 2. 5 mg/ml in human,the 5 min platelet maximum aggregation rate,compared with the control group(P〈0. 01),significantly decreased,the inhibition rate significantly increased,human PT、APTT,blood TT prolonged to different extent,compared with the control group(P〈0. 01),Fi B-C g/L content and AT-III activity significantly decreased(P〈0. 01),FDP mg/L content significantly increased(P〈0. 01),the difference was significant. Activities of coagulation factor II、V、VII、VIII、IX、X、XI、XII,compared with the control group,had different degrees of reduction(P〈0. 05). Conclusion: Xiyanping can decrease the platelet aggregation,activities of coagulation factor II、V、VII、VIII、IX、X、Ⅺ、XII in different degrees; prolong PT、APTT、TT; significantly increase FDP content; inhibit the endogenous and exogenous coagulation pathway to achieve anticoagulant effect,significantly decrease AT-III activity,can promote the activity of thrombin,play the procoagulant role,so Xiyanping has the dual effect of both
