波棱瓜子总木脂素对小鼠急性酒精性肝损伤的保护作用及机制探讨

摘　　要：目的:本实验为了研究藏药波棱瓜子总木脂素对小鼠急性酒精性肝损伤的保护作用及其机制。方法:用56度北京红星二锅头白酒建立小鼠急性酒精性肝损伤模型,用波棱瓜子总木脂素35mg/ml,25mg/ml和15mg/ml作为实验组,实验第31天,取肝脏,计算肝质量指数,测血清谷丙转氨酶(ALT)和谷草转氨酶(AST)活性,测定肝脏谷胱甘肽过氧化物酶(GSH-Px)、丙二醛(MDA)含量、黄嘌呤氧化酶(XOD)、超氧化物岐化酶(SOD)、Na+-K+-ATP酶并且进行肝脏病理学检查。结果:与模型组比较,波棱瓜子总木脂素25mg/kg降低小鼠血清AST和ALT的活力效果最为显著;波棱瓜子总木脂素35、25mg/kg提高小鼠肝脏Na+-K+-ATP酶、GSH-Px和SOD的活力,波棱瓜子总木脂素35、25mg/kg降低小鼠肝脏XOD和MDA的活力;HE显示各组肝细胞不同程度的损伤,部分肝细胞坏死,与模型组比较波棱瓜子总木脂素各组细胞破坏程度较轻。结论:波棱瓜子总木脂素对小鼠急性酒精性肝损伤具有保护作用,其机制与其提高机体抗氧化作用相关。Purpose：To discuss the protective effect and mechanism of total lignans from medicinal Herpetospermum seeds on alcohol-inducing acutehepatic injury in mice.Methods：Liver injury in mice was induced by 56°Beijing Red Star Erguotou wine,Doses（35mg/ml,25mg/ml和15mg/ml）of total lignans were used as the experimental groups.On the 31st day,the blood was taken from eyes,livers were collected andthe liver index was calculated.Meanwhile,levels of serum alanine aminotransferase（ALT）and aspartate aminotransferase（AST）.Contentsof glutathione peroxidase（GSH-Px）,malondialdehyde（MDA）,xanthine oxidase（XOD）,superoxide dismutase（SOD）and Na^＋-K^＋-ATPase in livers,meanwhile,hepatic pathological changes were examined.Results：Compared with the model group,total lignans（25mg/ml）decreased levels of ALT and AST（P〈0.05）,total lignans（35mg/ml,25mg/ml）increased levels of Na^＋-K^＋-ATP,GSH-Px and SOD intliver tissues（P〈0.05）and decreased levels of XOD and MDA in liver tissues（P〈0.05）,figures of histopathology examination showedthat different levels of injuries occurred in each group of total lignans.Conclusion：Total lignans can protect liver from acute alcoholic damageto some extent,the mechanism is related to improving the body＇s antioxidant effect.
