决明子水煎剂降低营养性高脂血症大鼠血脂量效关系研究

摘　　要：目的:研究决明子水煎剂对营养性高脂血模型大鼠血清血脂和肝脏组织中脂质的量效关系,为药食同源中药决明子的安全、有效、合理使用提供参考。方法:采用高脂饲料喂养的方法建立营养性高脂血大鼠模型,分别给予0.315、0.945、1.575、3.15、6.3 g/kg 5个决明子水煎剂剂量,通过检测不同剂量决明子水煎剂对动物血清中甘油三酯(TC)、总胆固醇(TG)、高密度脂蛋白(HDL-C)、低密度脂蛋白(LDL-C)、动脉粥样硬化指数(AI)5项指标及肝组织中TC、TG、丙二醛(MDA),总超氧化物歧化酶(T-SOD)项指标,经Graph Pad Prism 5.0软件拟合量效曲线,评价决明子水煎剂降低营养性高脂血症大鼠血脂量效关系,计算相关剂量阈参数。结果:决明子水煎剂对高脂血症大鼠血清血脂5项指标量效曲线呈良好的"S"型,剂量阈[D]20-[D]80和阈剂量[D]20多集中在0.599~4.108 g/kg范围内,肝脏组织中脂质4项指中有3项指标量效曲线呈良好的"S"型,剂量阈[D]20-[D]80和阈剂量[D]20多集中在0.639~1.795 g/kg范围内。结论:决明子水煎剂降低营养性高脂血模型大鼠血清血脂和肝脏组织中脂质量效曲线呈良好的"S"型,且其阈剂量均在0.599~0.639 g/kg,提示中剂量为有效剂量范围,折合成人体用量约为5.705~6.086 g,进一步验证后可为药食同源中药决明子的合理使用提供参考。Objective： To observe the dose-effect relationship of water decoction of cassiae semen on lowering lipid-in hyperlipidemia model rats and provide the reference for the safe,effective and rational use of the traditional Chinese medicine of cassiae semen. Method： The high fat feed was used to establish nutritional hyperlipidemia model rats and 5 cassiae semen water decoction groups（0. 315,0. 945,1. 575,3. 15,6. 3 g/kg） were set. And continuous gastric gavage was administered for 13 weeks. Five indexes of Total Cholesterol（TG）,High Density Lipoprotein（HDL-C）,Low Density Lipoprotein（LDL-C）,Atherosclerosis Index（AI） in serum and four indexes of TC,TG,malondialdehyde（MDA） and total superoxide dismutase（T-SOD） in liver tissues were measured on the 2 nd,5 th,8 th,11 th and 13 th week,respectively. After dose-response curves was fitted with Graph Pad Prism 6. 0 software,we evaluated the dose-response relationship of water decoction cassiae semen on lipid-lowering with hyperlipidemia model rats and calculated the relevant dose-threshold parameters. Results： The dose-effect curve of liver five indexes for hyperlipidemial rats was in well ＂ S＂ shape on Histology of serum blood,and the effective dose range [D]20-[D]80,[D]50 was at the rang of 0. 599 - 4. 108 g/kg. The dose-effect curve of four indexes for hyperlipidemiats was in well ＂ S＂ shapeon serum lipid levelsand the effective dose range [D]20-[D]80,[D]50 was at the range of 0. 639 - 1. 795 g/kg. Conclusion： Dose-effect Relationship of Cassiae Semen on water decoction Semen nutritional Hyperlipidemia for obtaining well ＂ S＂ shape of Histology of liver and serum lipid levels indexes was in the range of 0. 599 - 0. 639 g/kg,which indicated that the middle dosage range was effective,and the dose was equivalent to the clinical dosage of 5. 705 - 6. 086 g. The results will provide experimental evidence for the clinical medication of Cassiae Semen.
