摘  要:目的:制备四氢姜黄素(Tetrahydrocurcumin,THC)聚乙二醇-聚乳酸嵌段共聚物(MPEG-PLA)纳米颗粒(THC-MPEGPLA-NPs),考察其理化性质,探究四氢姜黄素纳米颗粒在体外对创面愈合的疗效。方法:采用溶剂萃取法制备THC-MPEG-PLANPs,利用透射电子显微镜观察纳米粒形态;激光粒度仪考察粒径和Zeta电位;超速离心法测定其包封率及载药量。利用皮肤成纤维细胞划痕实验模型探究纳米颗粒在体外的创面愈合作用。结果:制备的THC-MPEG-PLA-NPs外观呈圆形或类圆形,平均粒径为(32.70±1.15)nm,包封率和载药量分别为(99.49±0.03)%和(16.58±0.01)%;第12h,与未加入THC-NPs的成纤维细胞的迁移率56.3±8.5%相比,加入THC-NPs(THC给予量是5.0和10.00μg)的皮肤成纤维细胞的迁移率更高,分别是77.5±2.9%,69.3±4.7%。结论:溶剂萃取法可成功制备THC-MPEG-PLA-NP;四氢姜黄素聚乙二醇-聚乳酸嵌段共聚物纳米粒在创面愈合领域具有较大的临床应用潜力。Objective: To prepare Tetrahydrocurcumin-loaded MPEG-PLA nanoparticles( THC-NPs),to characterize their physicochemical properties,and to study the wound healing effect on Fibroblast cells in vitro. Methods: THC-NPs were prepared by solvent extraction method. We utilized the transmission electron microscope to observe the particle appearance,Zetasizer instrument to detect the diameter and Zeta potential,ultracentrifugation to determine entrapment rate and drug-loading rate. The fibroblasts migration assay was used to study its wound-healing in vitro. Results: We had successfully prepared THC-loaded MPEG-PLA nanoparticles. The optimal NPs were round with the nanometric size of( 32. 70 1. 15) nm,high entrapment rate of( 99. 49 0. 03) %,and drug-loading rate of( 16. 58 0. 01) %. At the 12 th hour,rates of the migration,77. 5 2. 9%( P 0. 05) and 69. 3 4. 7%( P 0. 01),were both high in the fibroblasts with THC-NPs( the feeding dosages of THC were 5. 0,10. 00μg,respectively),compared with that of the migration,56. 3 8. 5%,in the fibroblasts without THC-NPs. Conclusion:The solvent extraction method could be used to prepare THC-MPEG-PLA-NPs. We demonstrated that THC-NPs have the potential application in wound healing progress.
