摘  要:目的:探讨微寒中药玄参及其拆分组分对左甲状腺素钠片所致热证大鼠物质与能量代谢的影响。方法:除空白组外,各组分每日灌胃给予120mg/kg左甲状腺素钠片,造模1小时后灌胃给予玄参总提物水煎液及各拆分组分的水煎液,给药体积为10ml/kg,空白组和模型组给予等体积蒸馏水,连续灌胃15天后处死,测定肝脏葡萄糖激酶(GCK)、6-磷酸果糖激酶1(PFK-1)、丙酮酸激酶(PK)、丙酮酸脱氢酶(PDH)、乙酰辅酶A(Acetyl-Co A)、柠檬酸合成酶(CS)、异柠檬酸脱氢酶(ICD),肝脏和肌肉糖原合成酶激酶-3(GSK-3)、磷酸化酶(PYGL),肌肉中脂肪甘油三酯脂肪酶(ATGL),肝脏中琥珀酸-细胞色素C还原酶(CCR)、细胞色素C氧化酶(COX)、ATP合酶(ATPase)、腺苷激酶(ADK)。结果:与模型对照组相比,玄参总提取物1.350g/kg和大极性环烯醚萜1.205g/kg组GCK、PK、CS、ICD、CCR、COX和ADK值明显降低,玄参总提物1.350g/kg组肝脏中的PYGL含量明显降低,玄参总提物1.350g/kg、苯丙素提取物0.028g/kg组和大极性环烯醚萜1.205g/kg组的ATGL含量显著升高。结论:玄参对热证模型大鼠的物质与能量代谢有抑制作用。Objective: To discuss the effect of Scrophularia ningpoensis Hemsl on material and energy metabolism in pyretic syndrome rats induced by levothyroxine sodium. Methods: SD rats were administered with levothyroxine sodium solution at the dose of 120 mg/kg,and were administered with water decoction of Scrophularia ningpoensis Hemsl and varying split components 1 hour after rat models were made,the volume of the administration was 10 ml/kg. Rats were killed after 15 days,and contents of GCK,PFK-1,PK,PDH,Acetyl-CoA,CS and ICD in livers were assayed,contents of GSK-3 and PYGL in livers and muscles were assayed,the content of ATGL in muscle was assayed,contents of CCR,COX,ATPase and ADK in livers were assayed by Reagent kits. Results: Compared with the model group,contents of GCK,PK,CS,ICD,CCR,COX and ADK in serum were significantly decreased by large polar iridoid glycosides components and total composition components,PYGL in livers was decreased by total composition components,the content of ATGL in livers was increased by total composition components、large polar iridoid glycosides and phenylpropanoid glycosides components. Conclusion: Scrophularia ningpoensis Hemsl has good curative effects on pyretic syndrome rats.
