新益心宁神片镇静催眠作用的谱效关系研究

摘　　要：目的:建立新益心宁神片的指纹图谱,考察新益心宁神片对PCPA(对氯苯丙氨酸)大鼠失眠模型镇静催眠的效果,并分析其谱效关系。方法:建立不同批号新益心宁神片HPLC指纹图谱;取SPF级SD大鼠随机分为正常组、模型组、复方地西泮阳性对照组、各批号新益心宁神片组,采用腹腔注射PCPA建立大鼠失眠模型,正常组和模型组给予生理盐水,阳性对照组给予复方地西泮,其余各组给予不同批号的新益心宁神片,连续给药7 d。数据经统计分析后,用灰色关联分析谱效关系。结果:从新益心宁神片指纹图谱中共确定25个共有峰;不同批号新益心宁神片的镇静催眠效果存在显著性差异(P<0.05);灰色关联分析明确了11、18、21、12、14、6、19、24、7、10号色谱峰与镇静催眠作用相关性贡献较大。结论:通过谱-效关系研究,为探究新益心宁神片镇静催眠作用的有效物质奠定基础。Objective： To establish the fingerprint of New Yixin Ningshen tablet,to investigate the sedative and hypnotic effect of New Yixin Ningshen tablet on insomnia rat model induced by p-chlorophenylalanine（P-CPA）,and to analyze the spectrum-effect relasionship.Methods： Established different batches of New Yixin Ningshen tablet HPLC fingerprint.SD rats were randomly divided into normal group,model group,compound diazepam positive control group,different batches of New Yixin Ningshen tablet groups.The insomnia rats model was induced by abdominal cavity injection of P-CPA.Normal group and model group were given saline,the positive control group was given compound diazepam,and different batches number of New Yixin Ningshen tablet group were given extract of New Yixin Ningshen tablet continuous for 7 d.Then the grey relational analysis was used to investigate the spectrum-effect relationship.Results： 25 common peaks were detected in HPLC chromatography,different batches of New Yixin Ningshen tablet sedative hypnotic effect exist significant difference（P〈0.05）,the results showed that the sedative and hypnotic effect was mostly contributed to peak 11,18,21,12,14,6,19,24,7,10 of New Yixin Ningshen tablet after grey relational analysis.Conclusion： Through the spectrum-effect relationships research,lay the foundation for research of New Yixin Ningshen tablet sedative and hypnotic effect of effective material basis.
