马钱子苏木有效组分配伍对RA血瘀证模型大鼠炎症和血管内皮损伤的影响

摘　　要：目的：在类风湿关节炎（RA）血瘀证大鼠模型建立的基础上,观察马钱子、苏木有效组分（马苏组分）配伍对模型大鼠的关节炎症和血管内皮损伤的影响。方法：通过对RA血瘀证大鼠一般情况检查,足趾肿胀度及Elisa检测等方法分析各项指标,评估马苏组分配伍对大鼠炎症和血管内皮的影响。结果：马苏组分配伍能较好的改善RA血瘀证大鼠的一般状态,显著降低其足趾肿胀度,抑制炎症反应,改善血管内皮的损伤;降低模型大鼠血清中TNF-α、IL-1α、VEGF、COX-2、IL-1β、IL-6、IL-17、IL-15、VCAM-1、ICAM-1炎症因子水平,较好的逆转膝关节软骨的病理损伤改变,延缓关节软骨的退化进程。结论 ：马苏组分配伍能够有效抑制RA血瘀证模型大鼠的炎症及血管内皮损伤,延缓膝关节软骨退变,起到保护关节软骨的作用。Objective：To observe the effect of the combination of active ingredients from Maqianzi（Semen Strychni） and Sumu（Lignum Sappan） on joint inflammation and vascular endothelial injury in the rat model of rheumatic arthritis（RA） of blood stasis syndrome. Methods：According to the general condition of the rats,toe swelling degree,enzyme linked immunosorbent assay（ELISA） results,the effect of the combination of active ingredients from Semen Strychni and Lignum Sappan on joint inflammation and vascular endothelial injury was evaluated. Results：The combination of the active ingredients could improve the general condition of the rat model and significantly reduce the toe swelling degree,inhibit inflammation,relieve endothelial injury,reduce the serum level of inflammatory factors such as TNF-α,IL-1α,VEGF,COX-2,IL-1β,IL-6,IL-17,IL-15,VCAM-1,ICAM-1. Besides,the pathological changes of articular cartilage damage can be reversed,and the degeneration of articular cartilage can be delayed. Conclusion：The combination of the active ingredients from Semen Strychni and Lignum Sappan can effectively inhabit inflammation and vascular endothelia injury,protect joint cartilage to delay the degeneration of the knee-joint cartilage in the RA rat model of blood stasis syndrome.
