摘  要:目的研究GATA-4在对氯苯丙氨酸(4-Chloro-DL-phenylalanine,PCPA)抑制野百合碱(monocrotaline,MCT)诱导的大鼠肺动脉高压(pulmonary arterial hypertension,PAH)中的调节机制。方法雄性SD大鼠[体重(180±10)g]40只,随机分为对照组(control)、野百合碱组(MCT)、PAPC低剂量组(P1,腹腔注射PCPA 50mg/kg)、PCPA高剂量组(P2,100mg/kg)。21d后,检测肺动脉压力等血流动力学指标。HE染色观察肺血管重构。Western blot检测肺组织GATA-4及MMP-13蛋白表达。结果与对照组相比,MCT诱导大鼠的平均肺动脉压力、肺血管壁面积比显著增加,肺组织GATA-4及MMP-13蛋白表达明显增多。PCPA抑制肺血管重构,剂量依赖性地降低GATA-4及MMP-13的表达水平。结论GATA-4参与MCT诱导的大鼠PAH肺血管重构。色氨酸羟化酶抑制剂PCPA抑制大鼠PAH肺血管重构可能与调控GATA-4相关。Objective To study the effect of GATA-4 in the process of 4-Chloro-DL-phenylalanine(PCPA)inhibiting monocrotaline(MCT)-induced pulmonary artery hypertension(PAH).Methods The rats were divided into:control group,MCT group,P1 group(50 mg/kg intraperitoneal injection),P2 group(100 mg/kg intraperitoneal injection).21 days later,the pulmonary arterial pressure was measured.HE straining was used to observe the changes of pulmonary vascular remodeling,Western blot was used to detect the expression of GATA-4 and MMP-13 in rat lung tissues.Results The mean pulmonary artery pressure and the percentage of pulmonary vascular wall area,the expression levels of GATA-4 and MMP-13 proteins in rat lung tissues were increased significantly in MCT group than in control group,but decreased in PCPA group than in MCT group with dose-dependent manner.Conclusion GATA-4 involved in MCT-induced pulmonary arterial remodeling,the inhibition effect of PCPA on MCT-induced pulmonary arterial remodeling might be related to GATA-4.
