摘 要:目的:观察密蒙花颗粒剂对去势雄兔泪腺细胞肿瘤坏死因子α(TNF-α)、白细胞介素-1β(IL-1β)表达的影响,探究其治疗干眼症的机制。方法:将30只雄兔随机分为空白组、模型组、密蒙花颗粒剂组、安慰剂组和睾酮组。空白组除外均行双侧睾丸及附睾切除术建立干眼症模型。密蒙花颗粒剂组予密蒙花颗粒剂灌胃;安慰剂组予0.9%氯化钠溶液灌胃;睾酮组注射丙酸睾酮注射液。给药4周后免疫组化法检测泪腺细胞炎性因子TNF-α、IL-1β表达。结果:与空白组比较,模型组和安慰剂组TNF-α、IL-1β的表达显著升高(P<0.01);与模型组比较,密蒙花颗粒剂组和睾酮组TNF-α、IL-1β的表达显著降低(P<0.01)。结论:密蒙花颗粒剂可通过抑制泪腺细胞炎性因子TNF-α、IL-1β表达而治疗干眼症。Objective: To observe the effects of Buddleia Granules on the expression of TNF-α and IL-1β in lacrimal gland cells of castrated male rabbit. To explore the mechanism of treatment of dry eye. Methods: Thirty male rabbits were randomly divided into blank group, model group, Buddleia Granules group, placebo group and testosterone group. Except for the blank group, bilateral testicular and epididymal resection were performed to establish the dry eye model. The rabbits in the Buddleia Granules group were treated by the granules of Buddleia. The rabbits in the placebo group was intragastric administered with normal saline. The rabbits in the testosterone group was injected with Testosterone Propionate Injection. Four weeks after treatment, the expression of inflammatory factors TNF-α and IL-1β in lacrimal gland cells was detected by immunohistochemistry. Results: Compared with the blank group, the expression of TNF-α and IL-1β in model group and placebo group increased significantly(P0.01). Compared with the model group, Buddleia Granules group and testosterone group were significantly decreased(P0.01). Conclusion: Buddleia Granules can be used to treat dry eye by inhibiting the expression of inflammatory factors TNF-α and IL-1β in lacrimal gland cells.
