摘 要:目的:探讨梓醇配伍大黄酸对实验性自身免疫性脑脊髓炎(EAE)小鼠神经保护作用及髓鞘碱性蛋白(MBP)表达的影响。方法:C57BL/6j小鼠随机分为正常组、模型组、激素组、梓醇大黄酸组,以MOG35-55多肽免疫诱导产生EAE。HE染色观察炎性浸润情况,透射电镜观察髓鞘脱失及轴突损伤情况,免疫组化检测MBP的表达情况。结果:梓醇配伍大黄酸能够减少EAE小鼠外观行为改变率、降低死亡率,延长外观行为改变潜伏期;改善其临床症状和神经功能评分,在第20天和第40天,梓醇大黄酸组评分与模型组同期比较具有显著差异(P<0.05)。脑组织病理学检查结果显示:HE染色下各治疗组急性期和缓解期的炎性反应程度有所减轻;透射电镜下各治疗组髓鞘环形层状结构松散变形程度较模型组减轻;正常组MBP平均光密度值显著高于其他各组(P<0.05);急性期与缓解期,激素组、梓醇大黄酸组MBP平均光密度值显著高于模型组同期(P<0.05)。结论:梓醇配伍大黄酸能减轻EAE小鼠炎性反应,减轻EAE小鼠病理改变,缓解EAE小鼠髓鞘损伤。Objective: To investigate the effect of catalpa-rhein on the neuroprotection and the expression of myelin basic protein(MBP) in mice with experimental autoimmune encephalomyelitis(EAE). Methods: C57 BL/6 j mice were randomly divided into normal group, model group, hormone group and catalpa-rhein group. The EAE model was induced by myelin oligodendrocyte glycoprotein MOG35-55. The pathological changes in mice brains were observed with hematoxylin-eosin(HE) staining and transmission electron microscope. The expressions of myelin basic protein(MBP) in brains were measured by immunohistochemistry. Results: Catalpa-rhein could reduce the change rate of visual behavior, reduce death rate, prolong the incubation period of appearance behavior, improve the clinical symptoms and neurological functional scores. The histopathological examination result showed that the inflammatory reaction of second and remission stage decreased with HE staining; the loose deformation degree of the myelin sheath structure in the treatment group was less than that in the model group under transmission electron microscope; the average value of MBP in normal group was much higher than that in other groups(P〈0.05). In the acute phase, the mean serum level of MBP in hormone group and catalpol group was higher than that in model group(P〈0.05). In the remission stage, the MBP expression in the hormone group and catalpa-rhein group was higher than that in the model group(P〈0.05). Conclusion: Catalpa-rhein could reduce the inflammatory reaction, lighten the pathological changes, and relieve the myelin injury in EAE mice.
