芍药苷和芍药内酯苷对小鼠疼痛模型的镇痛作用及对β-EP、PGE_2的影响  被引量:120

摘  要:目的:研究赤芍、白芍及其有效成分芍药苷、芍药内酯苷对醋酸扭体法致小鼠疼痛模型的镇痛作用。方法:采用醋酸扭体法制备小鼠疼痛模型,灌胃给予赤芍水提物、白芍水提物,记录潜伏期和20min内扭体次数。进一步采用同一实验方法腹腔注射芍药苷和芍药内酯苷进行实验,并检测血清β-内啡肽(β-EP),大脑皮层β-EP、前列腺素E_2(PGE_2)含量。结果:与模型组比较,2g/kg赤芍水提物组潜伏期显著延长、扭体次数显著减少(P<0.01);1、0.5g/kg赤芍水提物组和2、1g/kg白芍水提物组扭体次数显著减少(P<0.01、P<0.05)。与1g/kg白芍水提物组比较,同剂量赤芍水提物组的扭体次数显著减少(P<0.05)。与模型组比较,各剂量芍药苷组和80mg/kg芍药内酯苷组的扭体次数均显著减少(P<0.01,P<0.05)。与40mg/kg芍药内酯苷组比较,同剂量芍药苷组的扭体次数显著减少(P<0.05)。与模型组比较,80mg/kg芍药苷组、80mg/kg芍药内酯苷组的血清和大脑皮层β-EP显著升高、大脑皮层PGE_2显著降低(P<0.01,P<0.05);40mg/kg芍药苷组仅血清β-EP显著升高(P<0.05)。结论:在醋酸扭体法致小鼠疼痛模型中,赤芍、白芍、芍药苷和芍药内酯苷均有镇痛作用,芍药苷为赤芍、白芍镇痛作用的共有成分;芍药内酯苷作为白芍的特征成分亦具有镇痛作用。芍药苷和芍药内酯苷镇痛作用的机制可能与升高血清和大脑皮层中β-EP水平、减少大脑皮层PGE_2生成或释放有关。Objective: To investigate the analgesic effect of paeoniflorin and albiflorin in acetic acid induced writhing mice. Methods: The model of acetic acid induced writhing mice was established, the mice received Radix Paeoniae Rubra and Radix Paeoniae Alba water by gavage, and the latency and 20 min writhing times were recorded. The paeoniflorin and albiflorin were used by intraperitoneal injection with the same experimental method. The serum β-EP and cerebral cortex β-EP and PGE_2 were also detected. Results: Compared with model group, 2 g/kg Radix Paeoniae Rubra could prolong latent phase and reduce the writhing times(P〈0.01); 1, 0.5 g/kg Radix Paeoniae Rubra, 2, 1 g/kg Radix Paeoniae Alba could reduce the writhing times(P〈0.01, P〈0.05). Compared with the same dose of Radix Paeoniae Alba, the writhing times of 1 g/kg Radix Paeoniae Rubra were obviously decreased(P〈0.05). Compared with model group, 80, 40, 20 mg/kg paeoniflorin and 80 mg/kg albiflorin could reduce writhing times(P〈0.01, P〈0.05). Compared with the same dose of albiflorin, the writhing times of 40 mg/kg paeoniflorin were obviously decreased(P〈0.05). Compared with model group, 80 mg/kg paeoniflorin and 80 mg/kg albiflorin could improve the levels of serum and cerebral cortex β-EP, and decrease the level of cerebral cortex PGE_2(P〈0.01, P〈0.05); 40 mg/kg paeoniflorin could increase the level of serum β-EP(P〈0.05). Conclusion: In the mice model of acetic acid-induced writhing, Radix Paeoniae Rubra and Radix Paeoniae Alba, paeoniflorin and albiflorin all had analgesic effects. Paeoniflorin is the common component of analgesic effect of Radix Paeoniae Rubra and Radix Paeoniae Alba, while albiflorin as a characteristic component of Radix Paeoniae Alba also has analgesic effect. The mechanism of paeoniflorin and albiflorin in the analgesic effect may be related to the increase of β-EP level in serum and cerebral cortex and the reduction or release of PGE_2 in cerebral cortex.
