肾虚质大鼠生长发育与海马蛋白激酶C表达的研究

摘　　要：目的:通过观察肾虚质大鼠生长发育情况及海马蛋白激酶C(PKC)含量变化,探讨二者的相关性及补肾中药的药理作用机制。方法:12周龄雌鼠受孕后,采用"猫吓鼠"造模法恐吓至其产子,仔鼠4周龄时继续恐吓,造成先天不足加后天失养复合型肾虚质仔鼠模型。继续恐吓仔鼠同时给予左、右归丸干预治疗;比较正常组、模型组与药物组生长发育情况并检测海马组织PKC含量。结果:(1)生长发育情况:仔鼠4月龄时,与正常组比较,肾虚质组体质量、脑重、肾重显著降低(P<0.05,P<0.01);左、右归丸组较肾虚质组有所改善(P<0.05);(2)海马PKC表达:与正常组比较,肾虚质组含量锐减(P<0.01);左、右归丸组较肾虚质组有显著提高(P<0.01)。结论:肾虚体质可致大鼠生长发育迟缓,其机制之一与海马组织中PKC含量的减少有关,同时补肾药物左、右归丸对其有改善治疗作用。Objective： By observing the growth and development as well as the protein kinase C（PKC） content in hippocampus of rats with kidney deficiency-consititution, to investigate the relationship between them and the pharmacological mechanism of kidney-tonifying Chinese medicine. Methods： Twelve week pregnant female rats were scared by cats to premature delivery, and newly born 4 week rats were scared with cats to establish congenital insufficiency pattern combining postnatal loss of nurture newborn rats with kidney deficiency constitution. When the intimidation of offspring began, Zuogui Pills and Yougui Pills were gave for treatment. Offspring rats＇ growth and development condition were observed, and hippocampus PKC expression were tested. Results：（1）Growth and development of offspring： when 4 months old, the weight of body, brain and kidney were decreased in the group with kidney-deficiency constitution（P〈0.01）, while the treatment of Zuogui Pills and Yougui Pills groups were better than those in the group with kidney-deficiency constitution（P〈0.05）.（2）Expression of PKC was decreased in the group of kidney-deficiency constitution, which were reversed by the treatment of Zuogui Pills and Yougui Pills groups（P〈0.01）. Conclusion： Kidney deficiency-constitution can cause growth retardation in rats. One of the mechanisms was related to the decrease of PKC content in hippocampus. At the same time, the kidney-tonifying Chinese medicine Zuogui Pills and Yougui Pills can improve the therapeutic effect.
