摘  要:目的探讨脐静脉内皮细胞(human umbilical vein endothelial cells,HUVECs)在川崎病(kawasaki disease,KD)并发冠状动脉损伤(coronary artery lesions,CALs)患儿血清作用下,核因子-κB(nuclear factor-kappa B,NF-κB)对基质金属蛋白酶9(matrix metalloproteases 9,MMP9)mRNA转录的影响。方法将HUVECs分为4组,分别为正常血清组(control组)、一般发热血清组(F组)、无冠脉损伤组(non-CALs组)和冠脉损伤组(CALs组)。免疫共沉淀(co-immunoprecipitation,ChIP)检测HUVECs中NF-κB p65与MMP-9启动子结合情况,RT-PCR检测HUVECs MMP-9mRNA水平。结果与对照组相比,经KD患儿,特别是并发CALs患儿血清作用后,HUVECs NF-κB p65可直接结合在MMP-9启动子上,同时MMP-9mRNA呈高表达。结论 HUVECs在KD并发CALs患儿血清作用下,NF-κB p65可直接结合在MMP-9启动子上促进其转录。Objective To investigate the relationship between nuclear factor-kappa B(NF-κB)and matrix metalloproteinase-9(MMP-9)mRNA in human umbilical vein endothelial cells(HUVECs)stimulated by the serum from children with coronary artery lesions of Kawasaki disease(KD).Methods HUVECs were cultured and were divided into 4 groups:normal serum group,general fever group,Non-CALs group and CALs group.Co-Immunoprecipitation(ChIP)was used to detect the relationship between NF-κB and MMP-9,and RT-PCR was used to detect the mRNA level of MMP-9.Results Compared with control groups,NF-κB p65 could bind the promoter of MMP-9 in HUVECs cultured with 10%serum from KD patients with coronary artery lesions.The mRNA level of MMP-9 was also up-regulated.Conclusion NF-κB p65 can promote the transcription of MMP-9 in HUVECs induced by the serum from KD patients with coronary artery lesions.
