摘  要:目的研究血小板微RNA(micro RNA)基因mi R-96-5p在血小板储存过程中抗凋亡的机制,探讨血小板储存时间的影响因素。方法用定量PCR方法检测储存起始和储存至第5天的健康血液捐献者的单采血小板中的24种与凋亡相关的mi RNAs的表达水平的变化,检测结果进行统计学分析,判断mi RNA在调节血小板存活或者凋亡方面的作用。结果与储存起始比较,血小板储存至第5天mi R-96-5p、mi R-16-5p、mi R-155-5p、mi R-148a-5p和mi R-296-5p的表达显著上升(P<0.01),而mi R-7-5p、mi R-145-5p、mi R-15a-5p、mi R-25-3p以及mi R-24-3p的表达显著下降(P<0.01)。转染mi R-96-5p抑制物至血小板72 h后,增加了细胞半胱氨酸蛋白酶-3(caspase-3)的活性,抑制了Bcl-2和Bcl-xl的表达,促进了Bax的积累和活性caspase-3的加工。结论 mi R-96-5p在血小板储存中可能发挥抗凋亡作用。Objective To study the anti-apoptosis mechanism of miR-96-Sp in platelet during the storage, and explore the factor affecting the platelet storage. Methods The expression changes of apoptosis related 24 miRNAs in apheresis platelets on the starting and 5 days of the storage was detected by quantitative real time PCR, and the results were sta- tistically analyzed. The effect of miRNAs on survival and apoptosis of platelet. Results Compared with the starting of the storage, the expression of miR-96-Sp, miR-16-Sp, miR-155-Sp, miR-148a-Sp and miR-296-Sp on 5 days of the platelet storage was significantly increased (P 〈 0.01), while the expression of miR-7-Sp, miR-145-Sp, miR-15a-Sp, miR-25-3p and miR-24-3p was significantly decreased (P 〈 0.01). 72 h after transfected the miR-96-Sp inhibitors into the platelet, the activity of caspase-3 was increased and the expression of Bcl-2 and Bcl-xl proteins was de- creased, the process of Bax accumulation and active of caspase-3 was promoted. Conclusion miR-96-Sp may play a rode in the anti-apoptosis of platele storage.
