摘  要:目的利用昆明小白鼠胚胎体外培养技术评估海淀区妇幼保健院新建实验室,选择合适的胚胎冷冻方法。方法(1)将小鼠体外和体内受精胚胎培养至第5天,观察囊胚形成率;(2)对第3天胚胎进行程序化及玻璃化冷冻和解冻。结果(1)两种方法的囊胚形成率均大于80%,且差异无统计学意义。(2)两种冷冻方法的胚胎复苏率均大于80%,玻璃化方法的胚胎复苏率高于程序化方法的胚胎复苏率,且差异有统计学意义。结论本实验中,小鼠体内、体外受精胚胎在体外均稳定发育,符合各项质控标准;实验结果还为本中心选择玻璃化方法冻存胚胎提供了依据。Objective:To assess the quality control for a newly constructed laboratory of Haidian maternal and child health hospital for in vitro embryo culture on experimental mouse basis of Kunming species. Methods:Embryos were cultured to examine the rate of blastocyst formation of in vitro and in vivo fertilization. Embryos were programmed or vitrification and thawing on the third day. Results:The blastocyst formation rates of the two methods were all greater than 80%,and the difference was not statistically significant. The embryo recovery rate of two methods were more than 80%,the recovery rate of embryo vitrification method is higher than the programmed method of the embryo recovery rate. Conclusion:In this experiment,the in vitro and in vitro fertilization embryos of Kunming mice developed stably in vitro,and met the requirements of various quality control standards. The experimental results also provide a basis for choosing vitrification to cryopreservation embryos in our center.
