左归丸含药血清对MC3T3-E1细胞分化过程中Cx43表达及GJIC功能的影响

摘　　要：目的：研究左归丸含药血清对培养MC3T3-E1细胞分化成熟过程中Cx43表达及GJIC功能的影响。方法：胰蛋白酶消化法传代培养MC3T3-E1细胞,通过血清药理学方法,设立正常对照组、左归丸组（5%、10%、15%）。分别用RT-PCR、Western blot法检测培养细胞第7、14、21天Cx43mRNA、蛋白表达;第21天划痕染料标记示踪法检测细胞GJIC功能。结果：随着培养时间增加,各组MC3T3-E1细胞Cx43mRNA、蛋白表达逐渐增强;GJIC染料标记检测中,可见绿色荧光在划痕两侧扩散,荧光定量分析存在组间差异。各浓度左归丸含药血清均可促进培养细胞的Cx43mRNA和蛋白表达、增强GJIC功能,并与含药浓度正性相关,其中以10%组作用最显著（P〈0.05或P〈0.01）。结论：左归丸含药血清能增强MC3T3-E1细胞Cx43mRNA、蛋白表达及GJIC功能,这可能是其促进MC3T3-E1细胞分化成熟的重要机制之一。Objective： To research the effect of Zuogui Pill medicated serum on the expression of Cx43 and GJIC function in cultured MC3T3-E1 cells differentiation and maturation process. Methods： MC3T3-E1 cells were cultured by trypsin digestion method,and the normal control group and Zuogui Pill group（5%,10%,15%） were established by serum pharmacological method. RT-PCR and Western blot method were used to detect the expression of Cx43 mRNA and protein on the 7 th,14 th and 21 st days. GJIC was assessed by scrape-loading dye transfer technique at day 21. Results： With the increase of culture time,the Cx43 mRNA and protein expression of MC3T3-E1 cells in each group were gradually increased,and the green fluorescence was spread on both sides of the scratches in GJIC function detection and there were differences between groups in fluorescence quantitative analysis. Each concentration of Zuogui Pill medicated serum can promote Cx43 mRNA and protein expression of MC3T3-E1 cells,and enhance the function of GJIC. The above results are positive related with the drug concentration,among which 10% had the most significant effect（P 0. 05 or P 0. 01）. Conclusion： Zuogui Pill serum can enhance Cx43 mRNA,protein expression and GJIC function of MC3T3-E1 cells,which may be one of the important mechanisms of Zuogui Pill promoting the differentiation and maturation of MC3T3-E1 cells.
