摘  要:目的:研究安神定志灵对TH、VMAT2的影响,从多巴胺合成、转运途径探讨安神定志灵治疗ADHD的作用机制。方法:依据随机数字将SHR大鼠分为模型组,西药组,安神定志灵低、中、高剂量组,每组8只,另设WKY大鼠8只设为正常组,分别灌胃4周。Percoll密度梯度离心法制备脑突触体,运用Western blot法检测TH、VMAT2的表达;运用PCR技术检测TH、VMAT2 mRNA表达水平,运用免疫组织化学技术检测TH、VMAT2阳性细胞表达量。结果:与正常组相比,模型组大鼠突触体内TH、VMAT2表达量显著降低(P〈0.01);经灌胃给药后,与模型组比较,西药组及安神定志灵中剂量组突触体内TH、VMAT2蛋白表达量、TH、VMAT2 mRNA表达量及阳性细胞数均显著上调(P〈0.01),安神定志灵低、高剂量组疗效不稳定。结论:安神定志灵能够显著改善SHR大鼠脑内TH、VMAT2的表达,影响脑内多巴胺的合成和转运,达到提高脑内多巴胺含量,控制ADHD临床症状的目的。Objective: To study the function of Anshen Dingzhi Ling on regulating TH and VMAT2,revealing the sedative effect mechanism of Anshen Dingzhi Ling in treatment of ADHD by regulating dopamine synthesis and transfer. Methods: Based on random number,SHR rats were divided into model group,western medicine group,Anshen Dingzhi Ling high,medium and low dose groups,each group 8,only setting up WKY rats 10 as the normal group. All rats were given administration by lavage for 4 weeks,respectively. Preparation brain synaptosome was prepared by Percoll density gradient centrifugation. Western blot method was used to detect the expressions of TH and VMAT2. PCR technique was used to detect TH and VMAT2 mRNA expression levels and immunofluorescence technique to detect TH and VMAT2 positive cells expressions. Results: Compared with normal group,model group's rats synaptic TH and VMAT2 expression levels were significantly reduced(P 0. 01). Compared with model group,western medicine group and Anshen Dingzhi Ling middle dose group's TH and VMAT2 in synapses and TH,VMAT2 mRNA expression quantity and number of positive cells were increased(P 0. 01). While,Anshen Dingzhi Ling low and high dose groups showed instable efficacy. Conclusion: Anshen Dingzhi Ling can significantly improve the expressions of TH and VMAT2 and affect the dopamine synthesis and transfer to achieve the aim of regulation the brain DA content to treat ADHD.
