摘 要:目的建立高效液相测定剑叶龙血树人工诱导结血物中龙血素A、B的方法,用于筛选有效的剑叶龙血树结血诱导剂。方法高效液相色谱法(HPLC)法,色谱柱安捷伦ZORBAX Eclipse XDB C18(4.6 mm×250 mm,5μm),流动相为乙腈-1%冰醋酸(40∶60),流速1.0 m L/min,柱温25℃,检测波长为280 nm。结果龙血素A在0.021 6~0.194 8μg范围内呈线性关系,回归方程为Y=-0.783 6X+1.825 1,r=0.999 8,龙血素B在0.020 1~0.181 1μg范围内呈线性关系,回归方程为Y=-0.478 5X+1.054 8,r=0.999 7,不同诱导剂诱导所得龙血素A含量在0.013%~0.099%,龙血素B含量在0.044%~0.097%。结论该方法快速、准确,可用于剑叶龙血树人工诱导结血物中龙血素A、B的检测,茉莉酸甲酯(50 mg/L)-苯甲酸(2 g/L)、硫酸镁(5%)-苯甲酸(2 g/L)、3吲哚丁酸(5 mg/L)-苯甲酸(2 g/L)、的诱导结血效果较好。A determination method of Loureirin A and Loureirin B in artificially induced dragon's blood of Dracaena cochinchinensis by HPLC was established to screen effective inducer. The Loureirin A and Loureirin B were separated on Agilent ZORBAX Eclipse XDB C18(4.6 mm×250 mm,5μm). UV detection wavelength was at 280 nm. The results indicated that the Loureirin A content showed good linear in the range of 0.021 6~0.194 8 μg,regression equation: Y=-0.783 6 X+1.825 1,correlation coefficient(r)=0.999 8;The Loureirin B content showed good linear in the range of 0.0201~0.1811 μg,regression equation: Y=-0.4785 X+1.054 8,correlation coefficient(r)=0.999 7.The content of Loureirin A ranged from 0.013% to 0.099% and the content of Loureirin B ranged from 0.044% to 0.097%.The determination method of Loureirin A and Loureirin B in artificially induced dragon's blood of Dracaena cochinchinensis is simple and repeatable, and can be used for screening the effective inducer of dragon's blood of Dracaena cochinchinensis.
