牛结核病刺激上清液在布病检疫中的应用

摘　　要：牛结核病和布病的检疫常称之为＂两病＂检疫。近年来牛结核病γ-干扰素酶联免疫吸附试验（ELISA）的应用日益增加,为了简化＂两病＂检疫,笔者尝试将牛结核病检测样品即牛型结核菌素（PPD）刺激上清液用于布病检测,用虎红平板凝集试验（RBT）、间接ELISA（i ELISA）试验和试纸条试验同时检测90头牛的血清样品和刺激上清液样品。结果表明：在RBT中,牛型PPD刺激上清液样品均呈非特异性阳性反应;在i ELISA试验中,血清样品的S/P值（样品OD值/阳性对照OD值×100%）平均值（194.84%）显著高于刺激上清液样品的平均值（183.57%,P〈0.05）,调整刺激上清液样品的判定标准后进行卡方检验,两种样品的结果差异不显著（P〉0.05）;在试纸条试验中,刺激上清液样品的阳性率（43.33%）显著大于血清样品的阳性率（28.89%,P〈0.05）。说明牛结核病γ-干扰素ELISA检测样品不能作为布病RBT和试纸条试验的样品,但可以作为布病i ELISA试验的样品。Bovine quarantine of brucellosis and tuberculosis is often called ＂two disease quarantine＂ in China. In recent years, the applicatioa of bovine IFN-γ ELISA test is increasing, in order to simplify the ＂two disease quarantine＂, the plasma stimulated by bovine PPD was used as sample to detect the antibody for brucellosis in this study. Sera and plasma of 90 cattle were detected by rose bangle test（RBT） , ELISA and strip test for brucellosis. Results showed that all plasma was false positive in RBT as the antigen self-agglutination. Also, in iELISA test, the average S/P value（ 194. 84% ）in sera was significantly higher than that of in plasma（ 183.57% ,P〈0. 05 ）. While the differentia was not significant by chi-square analyzing test（ P〉0. 05）after the cut-off standard was adjusted. In the strip test, the positive rate （43.33%） of plasma sample was significantly higher than that of in sera （ 28. 89% ,P〈0. 05）. It was illuminated that the plasma was not being used as RBT and strip testing samples, but can be tested as ELISA samples.
