摘 要:[目的]探讨红参皂苷提取物Rg3对1-甲基-4-苯基吡啶离子(MPP+)所致神经细胞氧化应激损伤的保护作用.[方法]取SH-SY5Y细胞株培养1d后加入1,5,10mg/L红参皂苷Rg3,4h后加入MPP+培养20h.利用MTT法检测细胞活性,采用流式细胞术检测线粒体膜电位(ΔΨm)及细胞内活性氧族(ROS)水平.[结果]与MPP+单独处理组比较,红参皂苷Rg3各剂量组的SH-SY5Y细胞损伤得到明显改善,ROS荧光强度值明显降低,ΔΨm荧光强度值明显升高,酪氨酸羟化酶表达水平明显升高(P<0.05).[结论]红参皂苷Rg3对MPP+诱导的细胞氧化应激具有保护作用.OBJECTIVE To explore the protective effects of red ginseng saponin Rg3 on oxidative stress damage of nerve cells induced by 1-methyl-4-phenylpyridinium ion(MPP+).METHODS1,5,10 mg/L red ginseng saponin Rg3 were added to the SH-SY5 Ycell lines which had already cultured for 1 day,4 hours later,MPP+was added for another 20 hours' culture.Cell viability was detected by MTT method,flow cytometry was used to detect the mitochondrial membrane potential(ΔΨ m)and cellular reactive oxygen species(ROS)levels.RESULTS Compared with the group treated only with 250 mmol/L MPP^+,in various dose of red ginseng saponin Rg3 groups,SH-SY5 Ycell damage was improved significantly,ROS fluorescence intensity was significantly reduced,ΔΨ m fluorescence intensity was significantly higher,the expression of tyrosine hydroxylase was obviously increased(P〈0.05).CONCLUSION Red ginseng saponin Rg3 had protective effects on cellular oxidative stress induced by MPP^+.
