摘 要:目的:研究尖叶假龙胆不同提取部位对胰岛素抵抗H4ⅡE细胞糖消耗的影响。方法:采用CCK-8法评价H4ⅡE细胞活性;高浓度胰岛素(10-6mol/L)诱导培养H4ⅡE细胞36 h,建立新的胰岛素抵抗细胞模型。葡萄糖氧化酶法检测尖叶假龙胆不同提取部位对胰岛素抵抗H4ⅡE细胞糖消耗的影响。结果:尖叶假龙胆不同提取部位浓度大于150μg/m L时,对H4ⅡE细胞活性均有明显的抑制作用。尖叶假龙胆不同提取部位中乙酸乙酯提取部位对胰岛素抵抗H4ⅡE细胞葡萄糖消耗影响显著,大孔树脂95%提取部位与大孔树脂70%提取部位也不同程度地影响胰岛素抵抗H4ⅡE细胞葡萄糖消耗,大孔树脂30%提取部位效果不明显。结论:尖叶假龙胆乙酸乙酯提取部位改善胰岛素抵抗效果最佳,大孔树脂95%提取部位与大孔树脂70%提取部位也可不同程度地改善胰岛素抵抗作用。Objective:To study the effects of different fractions from Gmtianella acuta influence on the glucose consumption of H4 Ⅱ E cells with insulin resistance.Methods:The H4 Ⅱ E cell activity was evaluated with method for the CCK-8.The H4ⅡE cells was cultured about 36 hours by insulin of high concentration(10-6mol/L) to establish new insulin resistance cell model.The effects of different fractions from Gmtianella acuta on the glucose consumption of H4ⅡE cells with insulin resistance was tested by glucose oxidase.Results:The effects of different fractions from Gmtianella acuta on H4 Ⅱ E cell activity were significantly inhibited,when their concentrations greater than 150 μg/m L.The ethyl acetate fraction of Gmtianella acuta significantly affected the glucose consumption of H4 Ⅱ E cells with insulin resistance.Macroporous resin-95% fraction and macroporous resin-70% fraction affected the glucose consumption of H4ⅡE cells with insulin resistance with different degrees,while macroporous resin-30% had no significant effects.Conclusion:The ethyl acetate fraction of Gmtianella acuta had best effects on insulin resistance,macroporous resin-95% fraction and macroporous resin-70% fraction could improve the insulin resistance with different degrees.
