摘  要:目的研究蛇床子素对胆管癌细胞的影响及其作用机制特点。方法采用MTT方法、Giemsa染色、Hoechst33258荧光染色、AO/EB荧光染色、流式细胞术和Western blot方法研究蛇床子素对胆管癌QBC939细胞增殖、形态变化,凋亡率、周期分布和凋亡相关蛋白表达变化的影响。结果蛇床子素对人胆管癌细胞QBC939的增殖有明显的抑制效果,且呈现剂量和时间依赖性,处理24 h和48 h的IC_(50)分别为0.169 63mmol/L和0.100 19 mmol/L;蛇床子素可诱导人胆管癌细胞QBC939的凋亡,0.15 mmol/L蛇床子素作用48 h后QBC939细胞凋亡率达到37.15%,并出现细胞凋亡的特征性形态变化,周期进程阻滞在G2/M期;蛇床子素可改变依赖于Caspase的凋亡途径中相关的蛋白Fas和Caspase-3的表达水平。结论蛇床子素具有良好的抗癌效果,依赖于Caspase的外源信号凋亡途径可能其凋亡机制之一。Objective To study the effects of Osthole on cholangiocarcinoma cells and its mechanism. Methods Using cell culture, MTT assay, Giemsa staining, Hoechst33258 fluorescence staining, AO/EB staining, flow cytometry and Western blot methods, the proliferation and morphological changes of cholangiocarcinoma cells, apoptosis rate, ex- pression change cycle and apoptosis related protein of cholangiocarcinoma QBC939 cells treated by osthole were investiga- ted. Results The results showed that proliferation of human cholangiocarcinoma QBC939 cells was inhibited by osthole with dose - and time -dependent manners, with the IC50 values of O. 169 63 mmoL/L and 0. 100 19 mmol/L for 24 h and 48 h, respectively. The apoptosis rate reached 37.15% after treatment with osthole for 48 h, and morphological changes in characteristics of cell apoptosis were detected. Meanwhile, the expression level of apoptosis - related proteins Fas and Caspase - 3 were changed after exposed to osthole. Conclusion Osthole has a good anti - cancer effect, which may be via the exogenous apoptosis pathway of caspase.
