摘  要:研究交泰丸中表小檗碱、药根碱、黄连碱、巴马汀、小檗碱在正常和慢性不可预见性温和应激(chronic unpredictable mild stress,CUMS)抑郁模型大鼠体内的药代动力学特性。根据Katz法建立CUMS大鼠抑郁模型,对正常大鼠和模型大鼠灌胃交泰丸混悬液,在不同时间点眼眶取血,采用LC-MS/MS法测定血浆中表小檗碱、药根碱、黄连碱、巴马汀、小檗碱的含量,运用DAS1.0软件计算药代动力学参数。结果表明,交泰丸中巴马汀、黄连碱、小檗碱、药根碱的C_(max)在抑郁大鼠体内分别为正常大鼠的1.99、2.14、2.3、1.82倍,AUC_(0-t)分别为1.23、1.25、1.29、1.46倍,AUC_(0-∞)分别为1.21、1.25、1.30、1.43倍,均具有极显著性差异。This study was aimed to explore the pharmacokinetics of epiberberine, jatrorrhizine, coptisine, palmatine, berberine of Jiaotai pill in the normal and depressed rats. According to ‘Katz' method, the model of chronic unpredictable mild stress(CUMS) was established. The extract of Jiaotai pill was orally administered to rats, and the blood samples were collected via the the oculi chorioideae vein according to the time schedule. The concentrations of epiberberine, jatrorrhizine, coptisine, palmatine, berberine in rat plasma were determined by LC-MS/MS, and the pharmacokinetic parameters were calculated by DAS1.0 software. Compared with normal rats, the C_(max) of palmatine, coptisine, berberine and jatrorrhizine in Jiaotai pill in depressed rats were 1.99, 2.14, 2.3, 1.82 times than the normal group, while the AUC_(0-t) were 1.23, 1.25, 1.29, 1.46 times and the AUC_(0-∞)were 1.21, 1.25, 1.30, 1.43 times, which were significantly different.
