摘  要:研究党参提取物对雪旺细胞增殖及相关蛋白表达的影响,为中药治疗周围神经损伤提供理论依据。将大鼠雪旺细胞株进行传代培养,通过MTT法筛选出党参提取物干预大鼠雪旺细胞生长的安全浓度。在安全浓度范围内,将雪旺细胞随机分为对照组和药物组,在药物组细胞培养板上加入不同浓度的党参提取物进行培养,分别在12、24、48、72和96 h观察其对细胞增殖的影响;用流式细胞术检测细胞周期及DNA含量百分率;用ELISA试剂盒检测雪旺细胞分泌神经细胞黏附分子(NCAM)及增殖细胞核抗原(PCNA)表达量的变化。结果显示,党参提取物干预大鼠雪旺细胞生长的安全浓度范围为250~4 000 mg/L;当党参提取物浓度为1 000 mg/L和2 000 mg/L时,与对照组相比,细胞增殖作用显著。结合MTT比色法及流式细胞仪检测结果表明,1 000 mg/L的党参提取物促进雪旺细胞增殖的作用极为显著。提示党参提取物在适当范围内可促进雪旺细胞增殖,以1 000 mg/L的效果最佳,并可上调雪旺细胞NCAM与PCNA的表达。To provide a theoretical basis for the treatment of peripheral nerve injury,the effects of Codonopsis extract on the expression of related proteins and proliferation of rat's Schwann cells were studied. The suitable concentration of Codonopsis extract for rat's Schwann cells growth was screened by MTT assay and rat Schwann cells were subcultured. Schwann cells were randomly divided into a control group and drug group in the appropriate concentration range,and the effects on cell proliferation were observed at 12 h,24 h,48 h,72 h and 96 h. The cell cycle and the percentage of DNA content were detected by flow cytometry; The expression of Schwann cells' secreting neural cell adhesion molecule(NCAM) and proliferating cell nuclear antigen(PCNA) were detected using ELISA kits detect. The results showed that Codonopsis extract appropriate concentration range of rat Schwann cells grown was 250-4 000 mg/L; compared with the control group,the cell proliferation was significant when the concentration of Codonopsis extract was 1 000 mg/L and 2 000 mg/L. The results of MTT colorimetric assay and flow cytometry showed that the proliferation of Schwann cells significantly when the concentration of Codonopsis extract was 1 000 mg/L. It is concluded Codonopsis extract in the appropriate range,especially 1 000 mg/L,can promote the proliferation of Schwann cells and up-regulate the expression of NCAM and PCNA.
