摘 要:目的用合成的缺位型硅钨酸盐Na_(10)[α-SiW_9O_(34)]·18H_2O(简称SiW_9)探究其体外抗肿瘤活性。方法通过IR光谱对其结构进行表征,并应用四甲基偶氮唑盐比色法分析SiW_9对人肝癌细胞(Hep G2)以及人肺癌细胞(A549)的增殖抑制活性。采用光学显微镜观察两种肿瘤细胞在不同浓度药物作用后的形态变化。结果缺位型硅钨酸盐SiW_9对两种肿瘤细胞均有增殖抑制作用,具有药物浓度依赖性,对两种肿瘤细胞的IC50值分别为(2.4±2.8)×10^(-4)mol/L和(1.8±3.1)×10^(-4)mol/L。光学显微镜下经药物作用后的肿瘤细胞出现皱缩、变圆、缩小等形态变化。结论硅钨酸盐SiW_9具有体外抗肿瘤活性,能够抑制肿瘤细胞增殖。Objective To synthesis lacunary silicotungstate Na10[α-SiW9O34]·18 H2O( abbreviated form: SiW9),and study its anti-tumor activity in vitro.Methods The structure of SiW9 was characterized by IR,and its inhibitory effect on the growth of human hepatoma HepG2 cells and lung cancer A549 cells was observed by MTT assay.The morphological change in tumor cells affected by different concentration of SiW9 was observed by optical microscopes.Results SiW9 could inhibit HepG2 and A549 cells proliferation in dose-dependent manner,with IC50 value of( 2.4 ± 2.8) × 10^-4 mol/L and( 1.8 ± 3.1) × 10^-4 mol/L,respectively.The optical microscope showed that the tumor cells appeared shriveled,rounded and reduced after treating with different concentrations of SiW9.Conclusion SiW9 could inhibit the proliferation of tumor cells.
