云南猪场隐孢子虫分子流行病学调查及种类鉴定

摘　　要：作为一种人兽共患的肠道原虫,隐孢子虫可感染包括猪在内的多种家畜和人类,但对云南省猪的隐孢子虫感染情况尚缺乏系统研究。为获悉云南省猪场隐孢子虫的感染情况及其虫种类型,本研究采集来自于云南四个代表性猪场的129份新鲜粪样,采用巢式PCR扩增猪隐孢子虫的18S rRNA基因序列,然后通过序列比对分析和构建进化树,解析隐孢子虫的种属关系;对不同地区及动物年龄的感染率进行风险因素分析。结果,有17个样本确定为隐孢子虫感染阳性,总体感染率为13.18%（17/129）,其中16例为成猪隐孢子虫,1例为猪隐孢子虫;风险因素分析显示,感染率在不同地区及不同的年龄段之间均有极显著的差异（P〈0.01）。本研究首次在云南省猪体内发现猪隐孢子虫虫种,研究结果为云南省猪隐孢子虫病的防控提供了基本数据。As a zoonotic intestinal protozoan,Cryptosporidium can infect a variety of animals including pigs,as well as humans.However,little is known of Cryptosporidium infection in pigs in Yunnan Province.The objective of the present study was to examine the prevalence and determine the species of Cryptosporidium in farmed pigs in Yunnan province.A total of 129 fresh fecal specimens were collected from four representative pig farms and tested by nested PCR amplification of the 18S rRNA gene.Then,the species of Cryptosporidium was determined by sequence comparison and phylogenetic analyses.The Cryptosporidium prevalence in different regions and ages of pigs were analyzed.Results showed that 17 samples were tested as Cryptosporidium positive,and the overall prevalence of Cryptosporidium was 13.18% （17/129）.16 were infected with C.scrofarum,1 was infected with C.suis.Analysis of risk factors showed that there was a significant difference among regions and age（P〈0.01）.This is the first report of the C.suis infection in pig in Yunnan province. The results of the present study provide the baseline data for preventing and controlling Cryptosporidium infection in pigs in Yunnan Province.
