摘 要:目的探讨以埃罗替尼为靶向基团的荧光探针在肺癌检测中的应用。方法制备以埃罗替尼为靶向基团的荧光探针。以肺癌A-549细胞作为实验组,宫颈癌Ca Ski、Si Ha和C33-A细胞作为对照组,分别应用0.1×10^(-6)、1×10^(-6)、10×10^(-6)mol·L^(-1)的荧光探针对肺癌A-549细胞及宫颈癌Ca Ski、Si Ha和C33-A细胞进行识别,荧光显微镜下观察以埃罗替尼为靶向基团的荧光探针对2组细胞的识别能力。结果探针浓度为10×10^(-6)mol·L^(-1)时,A-549细胞和Ca Ski细胞中均观察到强荧光信号,C33A和Si Ha细胞中几乎观察不到荧光信号;探针浓度为1×10^(-6)mol·L^(-1)时,A-549细胞中可观察到强的荧光信号,C33A中可观察到较弱的荧光信号,Ca Ski和Si Ha细胞中几乎观察不到荧光信号;探针浓度为0.1×10^(-6)mol·L^(-1)时,A-549细胞中可观察到强的荧光信号,Ca Ski、C33A和Si Ha细胞中几乎观察不到荧光信号。结论以埃罗替尼为靶向基团的荧光探针可特异性识别肺癌A-549细胞。Objective To explore the application of erlotinib based targeting fluorescent probe in the detection of lung cancer. Methods The erlotinib based targeting fluorescent probe was prepared. The lung cancer cells of A-549 were selected as experimental group,and cervical cancer cells of Ca Ski,Si Ha and C33-A were selected as control group. The A-549,Ca Ski,Si Ha and C33-A cells were identified by 0. 1 × 10-6,1 × 10-6,10 × 10^-6mol·L^-1 fluorescent probe; the identification ability of erlotinib based targeting fluorescent probe on cells in the two groups was observed under the inverted fluorescence microscope. Results When the concentration of fluorescent probe was 10 × 10^-6mol·L^-1,the strong fluorescence signal was observed in A-549 and Ca Ski cells,but the fluorescence signal was not observed almost in Si Ha and C33-A cells. When the concentration of fluorescent probe was 1 × 10^-6mol·L^-1,the strong fluorescence signal was observed in A-549 cells; the weak fluorescence signal was observed in C33 A cells; the fluorescence signal was not observed almost in Si Ha and C33-A cells.When the concentration of fluorescent probe was 0. 1 × 10^-6mol·L^-1,the strong fluorescence signal was observed in A-549 cells; the fluorescence signal was not observed almost in Ca Ski,Si Ha and C33-A cells. Conclusion Erlotinib based targeting fluorescent probe can specifically recognize lung cancer A-549 cells.
