摘  要:目的研究黄芩苷对人卵巢癌细胞株SKOV3、CAOV3增殖侵袭的作用机制。方法体外培养的人卵巢癌细胞株SKOV3、CAOV3分别与不同浓度的黄芩苷(0、5、7.5、10、12.5μmol/L)孵育24 h,用四甲基偶氮唑蓝(MTT)法检测细胞活性;用改良Boyden趋化小室法行癌细胞侵袭实验。卵巢癌细胞用不同浓度黄芩苷(0、5、7.5、10μmol/L)处理24 h,然后用蛋白质免疫印迹法(Western blot)和实时荧光定量聚合酶链反应(RT-PCR)实验检测基质金属蛋白酶2(MMP-2)蛋白水平表达及信使RNA(mRNA)水平。结果不同浓度黄芩苷对人卵巢癌细胞株SKOV3、CAOV3活性及侵袭均有抑制作用,且呈浓度依赖性;能明显降低MMP-2蛋白表达和mRNA表达,亦呈剂量依赖性。结论黄芩苷可在体外抑制人卵巢癌细胞株SKOV3、CAOV3活性及侵袭,其机制可能与抑制MMP-2活性有关。Objective To investigate the effect and mechanism of baicalin on the proliferation and invasion of human ovarian cancer cell line SKOV3 and CAOV3.Methods In vitro cultured human ovarian cancer cell lines SKOV3 and CAOV3 were incubated 24 h with different concentrations of baicalin(0,5,7.5,10,12.5 μmol/L),respectively.The cell viability was detected by four methyl azazolam blue(MTT),and the cell invasion assay was performed by modified Boyden chemokine chamber.Ovarian cancer cells were treated by different concentrations of baicalin(0,5,7.5,10 μmol/L) for 24 h.Then the expression levels of matrix metalloproteinase 2(MMP-2) protein and messenger RNA(mRNA) level were detected by Western blotting(Western blot) and real-time fluorescence quantitative PCR.Results Different concentrations of baicalin has inhibitory effects on the activity and invasion of human ovarian cancer cell line SKOV3 and CAOV3,and showed a concentration dependent manner,and it can significantly decrease the expression of MMP-2 and mRNA,and also showed a dose-dependent manner.Conclusion Baicalin can inhibit the activity and invasion of SKOV3 and CAOV3 cells in vitro,and its mechanism may be related to the inhibition of MMP-2 activity.
