摘 要:为研究脂肪酸去饱和酶基因FAD2在紫苏脂肪酸合成机制中发挥的重要作用,对紫苏FAD2基因及其编码的蛋白质序列进行了详细的生物信息学分析。结果表明,紫苏FAD2基因cDNA全长序列为1 545 bp,ORF为1 149 bp,共编码382个氨基酸残基;紫苏FAD2蛋白属于亲水性蛋白,具有6个典型的跨膜螺旋区,含有PLN02505保守结构域,属于Membrane-FADS-like超蛋白家族;系统进化树分析结果表明,紫苏与芡欧鼠尾草的亲缘关系较近,与红花较远。利用荧光定量PCR技术分析紫苏不同组织以及不同发育时期的种子中FAD2基因的表达特性,结果表明,紫苏FAD2在开花后20 d的种子中表达量最高。研究结果可为进一步阐明紫苏脂肪酸代谢机制奠定理论基础。To study the important role of fatty acid desaturase gene FAD2 in the synthesis of fatty acids in perilla seeds,in this study,the FAD2 gene and its encoding protein were analyzed in detail by bioinformatics.The results showed that the full length cDNA of FAD2 gene of Perilla frutescens was 1 545 bp,with an Open Reading Frame of 1 149 bp encoding 382 amino acids.The Perilla FAD2 protein belonged to the hydrophilic protein and had six typical transmembrane helix regions.It contained the conserved domain of PLN02505 and belonged to the Membrane-FADS-like superfamily.Phylogenetic tree analysis indicated that there was a closer genetic relationship between Perilla frutescens and Salvia hispanica,and there was a farther relationship with Carthamus tinctorius.The expression characteristics of FAD2 gene in different tissues of Perilla frutescens were analyzed by real-time PCR.The results showed that FAD2 reached the highest level in 20 days after flowering.The results provide a theoretical basis for further elucidating the metabolic mechanism of fatty acids in Perilla frutescens.
