摘  要:目的:通过丙硫氧嘧啶(PTU)造成大鼠甲状腺肿伴甲减的模型,观察贝牡莪消丸对甲状腺肿伴甲减大鼠TSH值及行为学方面的影响,进而探究该药物对本病的作用机制。方法:选取40只SD大鼠,随机选取其中10只作为空白组,其余30只给予丙硫氧嘧啶0.15 mg/g,每日1次,持续灌胃40天。造模成功后,将30只大鼠随机分为3组,即模型对照组、贝牡莪消丸组、优甲乐组,分别喂以生理盐水、贝牡莪消丸、优甲乐。给药30天后,以10天为1个周期,分别于给药前、给药10天后、给药20天后、给药30天后,进行大鼠行为学指标检测(敞箱实验)及新奇事物认知试验检测,并于给药30天后处死大鼠,镜下观察大鼠甲状腺细胞形态,并取血清测定TSH。结果:丙基硫氧嘧啶能够导致大鼠的甲状腺肿大并发甲减;贝牡莪消丸组的血清TSH值较优甲乐组显著降低;显著改善甲减大鼠异常空间认知能力。结论:PTU能够导致大鼠甲状腺肿大并发甲减,贝牡莪消丸可以促使甲状腺肿伴甲减大鼠TSH水平降低,并且显著改善甲减大鼠异常空间认知能力。Objective : To observe the effect of Beimu Exiao p i l on TSH and etliorats with hypothyroidism, and to further explore its action mechanism. Metliods : 10 SD rats were randomly se-lected from 40 rats as tlie blank control, the rest rats were given PTU(0. 15 mg/g) once per day for set up the model. The model rats were randomly divided into the model group, the Chinese medication ( BeimuE’xiao pill) group,and the western medication( Sodium Levotiiyroxine) group, with 10 rats in each group.Ethology test(open - field test) and novelty cognitive test were carried out in the rats before the treatment,10 d, 20 d, and 30 d after the treatment. Morphological changes of thyroid cells were observed and TSH value was measured 30 d after the treatment. Results: PTU could induce goiter concurrence withrats; TSH value was significantly decreased in the Chinese medication group compared medication group ; it can significantly improve the abnormal spatial cognition abiliConclusion: PTUcan lead to goiter concurrence with hypothyroidismin rats, Beimu Exiao decrease of TSH,it can significantly improve the abnormal spatial cognition ability in hypotliyroidism rats.
