三七总皂苷对血管紧张素Ⅱ致脐静脉内皮细胞损伤的保护作用

摘　　要：目的：研究三七总皂苷（血塞通）对血管紧张素Ⅱ（Ang Ⅱ）致脐静脉内皮细胞（HUVECS）损伤的保护作用。方法：体外培养HUVECS细胞,采用CCK8法,采用不同浓度的血管紧张素Ⅱ（Ang Ⅱ）干预正常HUVECS细胞,然后得到损伤条件为10^（-6） mol·L^（-1）,24 h。对于损伤后的HUVECS给予不同浓度的三七总皂苷（血塞通）进行保护治疗,设置100、50、25 mg·L-1为高、中、低剂量组,10^（-4）、10^（-5）、10^（-6）mol·L^（-1）氯沙坦为阳性对照组。按对照组、模型组、氯沙坦高、中、低剂量组,三七总皂苷高、中、低剂量组进行实验。对LDH、NO、NOS、eNOS、ET-1、PGI2、TX-B2、t-PA、PAI-1指标进行检测。结果：三七总皂苷对Ang Ⅱ损伤的细胞可以降低LDH、ET-1、TXB2、PAI-1的分泌,增加NO、NOS、eNOS、PGI2、t-PA的分泌。结论：三七总皂苷对Ang Ⅱ损伤的脐静脉内皮细胞具有保护作用,并可促进内皮细胞分泌扩血管物质和抗血栓物质。Objective： To study Panax notoginseng saponin（ PNS,XST） against prothrombotic effect induced by Ang Ⅱ and its protective effect on human umbilical vein endothelial cells（ HUVECS）. Methods： HUVECS cells were cultured in vitro,different concentrations of angiotensin Ⅱ（ Ang Ⅱ） were given,and found damaged condition was at 10^（-6） mol/L for 24 h. Different concentrations of PNS（ XST） were treated,then 100 mg/L,50 mg/L and 25 mg/L were used as high,medium and low dose groups. 10^（-4） mol/L,10^（-5） mol/L and 10^（-6） mol/L of losartan were used as positive control groups. LDH,ET-1,TX-B2,PAI-1,NO,NOS,eNOS,PGI2 and t-PA in cells were determined by ELISA. Results： PNS（ XST） could reduce LDH,ET-1,TX-B2,PAI-1 secretion and increase secretion of NO,NOS,eNOS,PGI2 and t-PA. Conclusions： PNS（ XST） might has anti-prothrombotic and protective effects on HUVECS injury induced by Ang Ⅱ.
