摘 要:目的:本课题拟通过建立痴呆型血管性认知功能障碍大鼠模型,观察脑络通颗粒对大鼠行为学以及脑组织GSH、NO、MDA含量及SOD、GSH-PX及NOS活性的影响,进一步揭示脑络通颗粒治疗痴呆型血管性认知功能障碍的作用机理。方法:应用双侧颈总动脉分期永久结扎法建立痴呆型血管性认知功能障碍大鼠模型,采用跳台法观察脑络通颗粒对模型大鼠行为学以及脑组织GSH、NO、MDA含量及SOD、GSH-PX及NOS活性等指标的影响。结果:跳台法显示,脑络通颗粒可以改善痴呆型血管性认知功能障碍大鼠模型的学习记忆能力。与模型组相比,尼莫地平组、中剂量组及高剂量组能升高大鼠脑组织GSH含量及SOD、GSH-PX活性,降低NO含量及MDA及NOS活性(P<0.01),而中剂量组及高剂量组在升高大鼠脑组织GSH含量及SOD、GSH-PX活性,降低NO含量及MDA、NOS活性与尼莫地平组比较无显著性差异(P>0.05)。结论:脑络通颗粒对痴呆型血管性认知功能障碍模型大鼠的学习记忆有明显的改善作用,可能与干预氧化应激反应有关。Objective: To observe the effect of Naoluo Tong granules on the behavior and the content of GSH, NO,MDA and the activities of SOD, GSH-PX and NOS in brain tissue of rats based on the vascular cognitive impairment of dementia type rat models. And further reveal the mechanism of action of Naoluo Tong granules in treating dementia vascular cognitive dysfunction. Methods: The model of dementia vascular cognitive impairment was established bypermanent ligation of bilateral common carotid arteries. The behavior of Naoluo Tong granules on the model rats and the contents of GSH, NO, MDA and SOD, GSH-PX and NOS activity and other indicators. Results: The jumping table method shows that Naoluotong Granules can improve the learning and memory ability of dementia vascular cognitive dysfunction rats. Compared with model group, nimodipine group, middle dose group and high dose group could increase GSH content, SOD, GSH-PX activity, NO content, MDA and NOS activity in rat brain(P0.01) Compared with nimodipine group, there was no significant difference between high dose group and high dose group in increasing GSH content, SOD, GSH-PX activity, decreasing NO content, MDA and NOS activity in brain tissue of rats(P0.05). Conclusion:Naoluo Tong Granules can significantly improve the learning and memory of rats with dementia vascular cognitive impairment, which may be related to the intervention of oxidative stress.
