摘 要:目的:研究益气活血药水提物与水提醇沉物对大鼠心肌细胞缺氧损伤的药效影响,并筛选出最佳的提取工艺。方法:建立体外心肌细胞缺氧模型,选取水提法及水提醇沉法制备的益气活血药进行干预,观察缺氧损伤后心肌细胞活力、细胞生存率、细胞形态学及上清中乳酸脱氢酶LDH释放量的变化情况,比较2种提取物的药效学差异。结果:益气活血药水提物与水提醇沉物均可一定程度上增加缺氧心肌细胞的活力,水提法益气活血药减轻LDH外漏优于水提醇沉法(P<0.01)。结论:益气活血药对H9c2心肌细胞缺氧损伤具有保护作用,且同剂量下以水提物为佳。Objective: The study aimed to observe the cardioprotective effects of different extracts of Yiqi Huoxue Decoction on cardiomyocyte hypoxia injury in vitro for optimum extracting technique selection. Methods:The hypoxia model was established for 24 h. The H9 c2 cells were treated with two different extracts of Yiqi Huoxue Decoction, including water extracts(WE) and alcohol precipitate(WEAP). The cell viability was examined by cell counting Kit-8(CCK-8). The changes of Morphological were observed by phase contrast microscope.The content of lactic dehydrogenase(LDH) was detected by biochemical kits. Results: The two different extracts of Yiqi Huoxue Decoction could significantly improve the cell viability. But WE was more effective than WEAP on the LDH levels(P〈0.01). Conclusion: Yiqi Huoxue Decoction has the protective effect on cardiomyocyte injury induced by hypoxia in H9 c2 cells. At the same dose, the effect of WE is superior to WEAP.
