摘 要:目的:探讨羚羊角与钩藤药对对发育期高热惊厥大鼠H_2S表达的影响。方法:筛选50只大鼠随机分为空白组、模型组、羚角组、钩藤组、羚羊角-钩藤组,每组10只,各药物组大鼠按10 m L/kg给予相应药液,空白组给予等容积蒸馏水,1次/d,连续21 d。给药60 min后,置于(45±0.5)℃恒温水浴箱热水浴实验,隔日1次,连续10次,末次给药24 h后,水合氯醛麻醉,经下腔静脉留取静脉血,离心,取血浆,-80℃冰箱保存,测H_2S含量。并取海马组织进行尼氏染色,观察病理变化。结果:羚羊角-钩藤组、羚羊角组及钩藤组在高热惊厥发生后海马CA1区神经细胞病理损伤程度有所减轻,其中羚羊角-钩藤组减轻更明显,优于羚羊角组、钩藤组。羚羊角组、钩藤组及羚羊角-钩藤组3组大鼠海马CA1区、CA3区的神经元数目明显多于模型组;而羚羊角组、钩藤组大鼠海马CA1区、CA3区的神经元数目少于羚羊角-钩藤组。羚羊角组、钩藤组及羚羊角-钩藤组3组大鼠的H_2S含量明显低于模型组,而羚羊角组、钩藤组H_2S含量高于羚羊角-钩藤组。结论:羚羊角与钩藤药对能减轻高热惊厥所致脑损伤,且药对优于单用,二者具有协同效应。Objective: To investigate the effect of Lingyangjiao(antelope horn) and Gouteng(uncaria) combined on the expression of H2 S in rats with febrile seizures. Methods: A total of 50 rats were randomly divided into model group, antelope horn group, uncaria group, antelope horn-uncaria group. The rats were given liquid medicine with 10 m L·kg-1 in each group, and the blank group was given the same volume of distilled water,once a day, for 21 days. After administration for 60 min, the rats were placed in(45±0.5) ℃ constant hot bath equipment every other day, for 10 times. After 24 h of the last drug administration, chloral hydrate anesthesia were applied and plasma of venous blood was collected by centrifugation and stored in-80 ℃, for further detection of H2 S. Pathologic diagnosis of Hippocampus was done by Nissl's staining. Results: Hippocampal CA1 neuronal cell injury in antelope horn-uncaria group, antelope horn group and uncaria group was significantly decreased, of which the antelope horn-uncaria group was more significant. The number of neurons in hippocampal CA1 and CA3 of rats in antelope horn group, uncaria group and antelope horn-uncaria group was significantly higher than that in model group, while the number of neurons in CA1 and CA3 of rats in antelope horn group and uncaria group was obviously lower than that in antelope horn-uncaria group. The levels of H2 S in antelope horn group, uncaria group and antelope horn-uncaria group were significantly lower than those in model group,while the levels of H2 S in antelope horn group and uncaria group were higher than those in antelope horn-uncaria group. Conclusion: Antelope horn and Uncaria combinational usage can reduce the brain injury caused by febrile seizures, which is better than single use, indicating the synergistic effect of these two drugs.
