大鼠腹腔异位心脏移植模型的建立与评价

摘　　要：目的建立一种稳定可行的大鼠腹腔同种异体异位心脏移植模型。方法选用40只SD大鼠作为供体和受体,随机分为A、B两组,在Ono创建的大鼠腹腔异位心脏移植模型的基础上,对麻醉方法、供心摘取、受体准备、吻合方法等进行了部分改进,术后统计手术时间及手术成功率。结果正式实验20例存活14例,存活率70%。术中吻合口出血致死1例;术中移植心脏复跳后停跳l例;术后24h死亡4例,供心阻断时间为(30±5)min,手术时间为(50±5)min。结论对经典的Ono大鼠腹腔异位心脏移植模型的手术步骤和操作流程进行优化和改进后,降低了建模的难度,缩短了手术时间,供体心脏保护好,手术成功率高。Objective To establish a simple and stable rat＇s abdominal heterotopic cardiac transplantation model with modified surgical technique.Methods 40 rats of heterotopic cardiac transplantation were performed using modified Ono＇s method, with modified technique of anesthesia, heart harvesting, recipient＇s vascular preparation and vascular anastomosis procedures. Donor＇s heart was transplanted to recipient’s abdominal cavity, the anastomosis were performed in an end-to-side manner between the donor＇s ascending aorta and the recipient＇s abdominal aorta, and between the donor＇s pulmonary artery and the recipient＇s inferior vena cava. The operation time and success rate were recorded.Results The average cold ischemia time was （30±5）minutes, total operation time was （50±5）minutes. In all 20 experiments, 14 recipient rats （70%） underwent successful heterotopic heart transplantation. One case died of hemorrhage, one died of cardiac systole, four cases died postoperatively.Conclusion Using modi？ed Ono＇s procedures can increase the success rate of rat cardiac transplantation and make the operation easier.
