摘  要:【目的】从转录水平整体分析狗牙根在水淹生境下的基因表达模式对探索植物的抗水淹机制具有重要意义。【方法】本研究通过转录组测序研究了水淹(处理组)和非水淹(对照组)生境下的三峡库区消落带适生狗牙根的转录因子(transcription factor,TF)基因表达变化。【结果】测序数据经拼接共获得147 256条非冗余Unigenes,经Nr、Nt、Swissprot、KEGG、COG、Interpro和GO数据库可注释72.84%的Unigene;筛选到与狗牙根响应水淹胁迫有关的54个家族的785条差异表达TF,其中属于27个家族的348条TF上调表达,34个家族的437条TF下调表达;WRKY、b HLH、MYB、AP2-EREBP、MYB-related、Trihelix、NAC、MADS和TCP等是9个差异表达TF数量最多和表达量变化最显著的TF家族。基于实时定量PCR对水淹生境下狗牙根TF表达水平进行了检测,结果与转录组分析基本一致。【结论】初步分析了水淹生境下狗牙根转录因子的特征表达谱,为进一步筛选与抗水淹有关的关键基因和调控基因提供基础。【Objective】It is of great significance to explore the mechanism of plant's resistance to water flooding from the transcription level by overall analysis of the gene expression pattern of Cynodon dactylon under waterlogging.【Method】Gene expressions of transcription factor of Cynodon dactylon in Three Gorges Reservoir area under flooding( treatment group) and non-looded( control) group were studied based on transcriptome analysis. 【Result】147 256 pieces of non-redundant Unigenes were obtained after assembly,and 72. 84 % of Unigenes numbers was annotated by database of Nr,Nt,Swissprot,KEGG,COG,Interpro and GO. 2928 pieces of transcription factors belonging to 62 families were predicted by alignment in database of transcription factor domains. 785 pieces of differentially expressed transcription factor of54 families were screened,associating with Cynodon dactylon in response to flooding stress,and among them,348 transcription factors belonging to 27 families were up-regulated,and the 437 transcription factors of 34 families was down-regulated. WRKY,b HLH,MYB,AP2-EREBP,MYB-related,Trihelix,NAC,MADS and TCP were the largest number of differentially expressed transcription factors and the most significant changes in expression. Based on real-time quantitative PCR,the expression level of transcription factors of Cynodon dactylon under waterlodding was detected,and the results were consistent with those of transcriptome analysis. 【Conclusion】The characteristic expression maps of the transcription factor of Cynodon dactylon under waterlogging was initially analyzed,which provided a basis for screening the key genes and regulatory genes associated with waterlogging resistance.
