摘 要:【目的】为研究POU1F1基因3种剪切体在藏鸡不同的阶段中对藏鸡生长的影响。【方法】以藏鸡为研究对象,采用荧光定量技术,对POU1F1 3种剪接体在藏鸡的4个时期(81、119、154、210 d)垂体、腿肌和胸肌中的表达量进行检测,并与其生长性能进行关联分析。【结果】POU1F1基因的3个剪接体在4个时期中均于119日龄时有一较高值,之后随日龄增加而降低;且表达水平POU1F1-β>POU1F1-α>POU1F1-ω;在垂体、胸肌和腿肌中,垂体中表达远高于其他2个组织;在垂体中,雌性的表达量高于雄性。藏鸡POU1F1-α的表达量与活体重、半净膛重、全净膛重和腿肌重4个指标呈显著相关,与屠体重呈极显著相关;119日龄时,POU1F1-α和POU1F1-ω均与活体重、屠体重、半净膛重和腿肌重呈显著相关;154日龄时,POU1F1-ω与参与分析的性状没有显著相关性。【结论】POU1F1基因的3种剪接体在119日龄之前对鸡的生长发育影响最为显著,POU1F1-α和POU1F1-β对藏鸡的生长发育均有显著影响,可作为影响生长发育的候选基因,对提高家畜生长速度,增加畜产品的产出能力提供理论基础。【Objective】Effects of POU1 F1 gene in three kinds of shear in Tibetan chicken in the different stages on the growth of the Tibetan chicken was studied.【Methods】Taken Tibetan chicken as the research object,using fluorescence quantitative technology,the expression amounts of the POU1 F1 three spliceosome of the pituitary,leg muscle and chest muscle in Tibetan chicken four growth periods(81,119,154,210 days) were determined,and its association with growth traits was analyzed.【Results】The splicing variants of POU1 F1 gene all had a high expression level at 119 days in four periods and then decreased with increasing age; And their expression levels were in the order of POU1 F1-β POU1 F1-α POU1 F1-ω; Among the pituitary,chest and leg muscle,the expression in pituitary was much higher than that of other two groups; In the pituitary gland,the expression of female was higher than that of male. The expression of POU1 F1-α with 4 indicators( live weight,semi eviscerated weight,eviscerated weight and leg muscle weight) were significantly correlated and highly significantly correlated with the carcass weight; At the age of 119 days,POU1 F1-α and POU1 F1-ω were significantly correlated with the live weight,carcass weight,half eviscerated weight and leg muscle weight; At the age of 154 days,there was no significant correlation between POU1 F1-ωand involved in the analysis of the characters.【Conclusion】The three spliceosome POU1 F1 gene before 119 days of age affecting on the growth and development of the chicken was the most significant,and POU1 F1-α and POU1 F1-β had a significant effect on the growth and development of Tibetan chicken,which could be used as candidate genes to improve livestock growth,increase the output capacity and provide a theoretical basis for livestock.
