摘　　要：目的 :观察角质细胞生长因子 (KGF)在体外对常染色体显性遗传型多囊肾病 (ADPKD)囊肿衬里上皮细胞的促增殖作用并探讨其可能的作用机制。方法 :外源性给予不同浓度的 KGF(0～ 10 0 ng/ ml) ,采用 MTT法检测细胞增殖情况 ,流式细胞术检测细胞周期 ,在电镜下观察细胞形态。结果 :KGF呈剂量、时间依赖性地促进了 ADPKD囊肿衬里上皮细胞的增生 ,表现为细胞数量增多 ,S期细胞比率增高 ,细胞分裂活跃 ,增生的细胞形态尤其是细胞核较正常体积增大。结论 :KGF可显著刺激 ADPKD囊肿衬里上皮细胞的增殖 ,提示 KGF可能参与 ADPKD肾囊肿的形成和发展。Objective:To observe the effect of keratinocyte growth factor (KGF) on proliferation of autosomal dominant polycystic kidney disease (ADPKD) cyst lining epitheliar cells in culture. Methods:ADPKD cyst lining epitheliar cells were incubated with different concentration of KGF,ranging from 0 100 ng/ml.Cell proliferation was assessed by MTT colorimetric assay.Cell cycle was detected by flow cytometry and morphologic change was evaluated by electron microscopy. Results:After administration of KGF, the typical change of ADPKD cyst lining epitheliar cells proliferation was in a dose and time dependent manner, reflected by increase of MTT value and the ratio of S phase in cell cycle and ultrastructural changes revealed by transmission electron microscopy. Conclusion:KGF significantly stimulates the proliferation of ADPKD cyst lining epitheliar cells, which may be involved in the formation and development of renal cyst in ADPKD.
