摘  要:本研究以裂叶垂枝桦木质化、未木质化茎段为外植体,对取材时期、消毒时间、激素浓度及外植体类型等影响因素进行了系统的研究。结果表明:4月中旬为裂叶垂枝桦外植体取材的最佳时期;选用0.1%Hg Cl2为消毒试剂,裂叶垂枝桦木质化茎段最佳消毒时间为10 min,幼嫩茎段最佳消毒时间为3 min;裂叶垂枝桦幼嫩茎段诱导最适培养基和激素浓度配比为:MS+0.5 mg/L 6-BA+0.05 mg/L NAA+0.2 mg/L GA_3+6 g/L琼脂+20 g/L蔗糖,其分化率可达83.3%;木质化茎段离体诱导最适培养基和激素浓度配比为:MS+0.2 mg/L6-BA+0.2 mg/L NAA+0.2 mg/L GA_3+6 g/L琼脂+20 g/L蔗糖,分化率可达73.3%。In this research,we took lignified stems and not lignified stems of Betula pendula as explants,and the influences of inoculating periods,disinfection time,hormone concentrations and explant types were studied systematically.The results showed that the middle of April was the best period for explant inoculating of Betula pendula.The HgCl2 with 0.1% concentration was used as disinfectant,and the optimal time of disinfection for lignified stems and not lignified stems were 10 min and 3 min,respectively.The ratio of the optimum medium and hormone concentration induced young stems segments of Betula pendula was MS +0.5 mg/L BA +0.05 mg/L NAA+0.2 mg/L GA3+6 g/L agar+20 g/L sucrose,and the differentiation rate could reached 83.3%; The ratio of the optimum medium and hormone concentration induced lignification stem segments in vitro was MS +0.2 mg/L BA+0.2 mg/L NAA+0.2 mg/L GA3+6 g/L agar+20 g/L sucrose,and the differentiation rate could reached 73.3%.
