摘 要:杨山牡丹‘凤丹’(Paeonia ostii T.Hong et J.X.Zhang var.lishizhenii B.A.Shen)抗逆性强,适应性广,主要用作观赏、药用和新型油料作物。‘凤丹’种子繁殖的个体差异大,组织培养不定芽增殖系数低,生根困难,移栽驯化难以成活,严重制约良种繁育和新品种培育。本研究以‘凤丹’鳞芽探究鳞芽表面消毒最佳方式;生长调节剂、蔗糖浓度、继代周期、防褐化剂对不定芽扩繁的影响;不定芽珍珠岩基质容器化扦插生根、活性炭(AC)-吲哚丁酸(IBA)集成诱导不定根发生和移栽驯化。初步建立组织培养工厂化育苗技术体系,鳞芽以有效氯1%的二氯异氰尿酸钠水溶液处理20 min,去污染成活率高达66.0%;不定芽在6-苄氨基嘌呤(6-BA)0.5 mg/L、吲哚-3-乙酸(IAA)0.05 mg/L的改良WPM培养基上增殖系数最高达2.73;蔗糖浓度为20 g/L、继代周期为35 d有利于不定芽分化;培养基中添加维生素C(VC)15 mg/L、核黄素(VB2)20 mg/L抗褐化效果最佳。AC-IBA集成诱导不定芽生根,AC 0.5 g/L、AC培养时间为15 d、IBA 15 mg/L诱导生根率53.3%,小植株移栽驯化成活率26.7%;不定芽珍珠岩基质容器化扦插,IBA 20 mg/L诱导生根率40%,驯化成活率97.8%。优化的‘凤丹’组织培养体系为工厂化良种繁育奠定技术基础。Paeonia ostii va r.lishizheni has strong resistance to stress and wide adaptability,mainly used as ornamental,medicinal and new oil plant.While,the plant individual difference of seed propagation is large,the in vitro shoot propagation coefficient bud is low,the rooting is difficult,and the transplanting and hardening are difficult to survive.All these seriously restrict the plant tissue culture techniques used in micropropagation and improvement.In this research,the buds of P.ostii var.lishizhenii were used to research;The effect of aseptic culture,plant growth regulator cooperation,sucrose concentrations,subculture intervals and anti-browning agents on the shoot multiplication.Container cutting on perlite matrix and activated carbon(AC)-beta-indolebutyric acid(IBA)integrated induced rooting,transplantation and acclimatization.The preliminary industrialized propagation technique system of tissue culture was established.The result showed that the survival rate of buds reached 66.0%with 1% effective chlorine of dichloroisocyanuric acid sterilizing 20 min;the multiplication coefficient could reach2.73 on the modified WPM medium containing 6-benzylaminopurine(6-BA) 0.5 mg/L and indole-3-acetic acid(IAA) 0.05 mg/L;sucrose 20 g/L and subculture interval 35 d were beneficial for the multiplication of shoots.Vitamin C(VC) 15 mg/L and Vitamin B2(VB2) 20 mg/L added to the medium was optimal for the cultures' antibrowning.With the AC-IBA integrated induced rooting method,the rooting rate of shoots was 53.3% as AC 0.5 g/L culturing for 15 d and IBA 15 mg/L.After transplantation and acclimatization,the plantlets survival rate reached26.7% IBA 20 mg/L improved the rooting rate of shoots to 40% with the container cutting rooting on perlite matrix method,and the rooting plantlets survived at 97.8% though acclimatization.The modified tissue culture system lays on the technique foundation for industrialized micropropagation of improved P.ostii var.lishizhenii.
