摘 要:目的:观察四妙方加味对2型糖尿病模型(T2DM)小鼠血糖、血脂及肝脏组织病理的影响及对慢性脂肪肝模型大鼠血脂、肝指数、Lee’s指数、脂肪指数的影响。方法:①通过腹腔注射链脲佐菌素(STZ)建立,T2DM小鼠模型,选取成模小鼠随机分为6组:对照组,模型组,四妙方加味低、中、高剂量组和二甲双胍组,另设空白对照组。每天灌胃给予相应药物或蒸馏水,3周后测小鼠血糖及血清中总胆固醇(TC)、三酰甘油(TG)含量,计算肝重指数并观察小鼠肝组织病理变化。(④灌胃给予高脂乳剂建立慢性脂肪肝大鼠模型,造模成功的大鼠随机分组5组,即模型组,阿托伐他汀组,四妙方低、中、高剂量组。另设空白对照组。各组口服给予相应的药物或蒸馏水。8周后取血测血清中TC、TG含量,称取大鼠脂肪的重量、肝脏重量,计算Lee’s指数和脂肪指数。结果:①与模型组比较,二甲双胍组和四妙方加味低、中、高剂量组小鼠血糖含量均显著下降(P〈0.05或P〈0.01);四妙方加味中、高剂量组TC含量明显低于模型组(P〈0.05或P〈0.01);四妙方加味各组TG含量均显著下降(P〈0.05或P〈0.01);二甲双胍组和四妙方加味比较,二者血糖、血脂、肝指数水平上无明显差异(P〉0.05)。与模型组比较,四妙方加味中、高剂量组,均未发生脂肪变性和重度水样变性,脂肪变性指标差异不明显,水样变性指标有显著差异(P〈0.01)。与二甲双胍相比,四妙方加味对肝组织的损伤有较强的缓解作用。(④与对照组比较,四妙方加味中、高剂量组及阿托伐他汀组能降低血清中TC、TG含量(P〈0.05或P〈0.01)。四妙方加味低剂量组大鼠与模型组大鼠比较,脂肪指数降低(P〈0.05);中、高剂量及阿托伐他汀组大鼠与模型组大鼠比较,肝指数、Lee’s�Objective :To explore the effect of modified Simiao Granule on blood glucose,blood lipid,and liver tissue in mouse with type 2 diabetic,and on the blood lipid,liver index , Lee's index and fat index in rat with chronic fatty liver. Method: (1) Type 2 diabetic mouse model was established by intraperitoneal injection of streptozotoein(100 mg/kg). After the models were successfully established,mice were randomly divided into 6 groups:control group,model group,Simiao Granule low dose group,Simiao Granule middle dose group,Simiao Granule high dose group,and metformin group. In each group,mice were given intragastric administration of the corresponding drugs or distilled water daily. After 3 weeks,the levels of total cholesterol (TC) and triglyceride(TG) in serum and blood glucose were measured. The liver weight index of mice was calculated and the histopathological changes were observed under optical microscope. (2) High fat emulsion was given by gavage to build chronic fatty liver rat model,and then the model rats were randomly divided into 5 groups:model group,atorvastatin group,Simiao Granule low dose group,Simiao Granule middle dose group,and Simiao Granule high dose group. A blank control group was built up beside the above groups. Each group drugs or the was orally given the corresponding same volume of distilled water. Eight weeks later,levels of TC and TG in the serum were measured. Body fat weight and liver weight were measured. The Lee's index and the fat index were calculated. Results: (1) Compared with the model group,the fasting blood glucose level significantly decreased in the metformin group and modified Simiao Granule of low,middle and high dose groups(P〈0.05 or P〈0.01). The level of TC in the Simiao Granule of middle and high dose group was significantly lower than that in the model group(P〈0.05 or P〈0.01).
