摘 要:[目的]建立同时检测新麦纤散中5种成分:三七皂苷R1、人参皂苷Rb1、Coelonin、薯蓣皂苷和茯苓酸含量的方法。[方法]采用反相高效液相色谱(reversed phase-high performance liquid chromatography,RP-HPLC)梯度洗脱法测定三七皂苷R1、人参皂苷Rb1、Coelonin、薯蓣皂苷、茯苓酸的含量。色谱条件:phenomenex-C18色谱柱(250mm×4.6mm,5μm),流动相乙腈和0.1%磷酸水梯度洗脱,流速1.0 m L·min^(-1),柱温30℃,检测波长为203nm。[结果]三七皂苷R1、人参皂苷Rb1、Coelonin、薯蓣皂苷、茯苓酸分别在0.65~16.25μg·mL^(-1)(r^2=0.9996),0.42~10.50μg·mL^(-1)(r^2=0.9997),0.08~2.00μg·mL^(-1)(r^2=0.9998),0.37~9.25μg·mL^(-1)(r^2=0.9986),0.16~4.00μg·mL^(-1)(r^2=0.9999)范围内线性关系良好,平均加样回收率分别为98.53%、99.02%、98.55%、95.43%、97.66%,相对标准偏差(relative standard deviation,RSD)分别为1.27%、0.54%、0.53%、1.48%、0.81%。[结论]RP-HPLC梯度洗脱法操作简便,结果准确,精密度高,重现性好,能同时测定新麦纤散中三七皂苷R1、人参皂苷Rb1、Coelonin、薯蓣皂苷、茯苓酸的含量,可为完善新麦纤散的质量控制方法提供科学的依据。[Objective] To establish the method for simultaneous determination of panax notoginseng saponins R1, ginsenoside Rbl, Coelonin, diosgenin and poria acid in New Maixiansan. [Methods] The contents were determined by reversed phase-high performance liquid chromatography(RP-HPLC) gradient elution. Chromatographic conditions: phenomenex-C 18 column (250mm-4.6mm, 5μm), mobile phase acetonitrile and 0.1% phosphoric acid gradient elution, flow rate 1.0mL·min^-1, column temperature of 30℃, detection wave length of 203nm. [Results] Panax notoginseng saponins R1, ginsenoside Rbl, Coelonin, diosgenin and poria acid presented better linear relationship in the ranges between 0.65-16.25μg'mL^-1 (r2=0.9996),0.42-10.50μg.mL^-1 (r^2=0.9997),0.08- 2.00μg·mL^-1 (r^2=0.9998),0.37-9.25μg·mL^-1 (r2=0.9986),0.16-4.00μg·mL^-1(r2=0.9999). The average recoveries were 98.53%, 99.02%,98.55%, 95.43% and 97.66% respectively. RSDs were 1.27%, 0.54%, 0.53%, 1.48%, 0.81%. [Conclusion] The method, easy to operate, accurate results, high precision and good reproducibility, could be used to determine the contents of panax notoginseng saponins R1, ginsenoside Rbl, Coelonin ,diosgenin and poria acid in new Maixiansan simuhaneously and it could orovide scientific evidence for improving aualitv control method of new Maixiansan.
