三种免核酸提取PCR试剂盒检测乙肝患者血清HBV-DNA的临床对比研究

摘　　要：目的针对乙型肝炎病毒的检测,选取3种免核酸提取PCR试剂盒,研究其临床应用价值。方法采集24例乙型肝炎患者血清以及24例阴性血清标本,分别用3种免核酸提取PCR试剂盒进行检测,对比这3种试剂盒的检测灵敏度以及特异性,检测结果以荧光定量PCR结果为标准。结果针对24例乙型肝炎患者血清检测,结果发现,3种免核酸提取试剂盒中,购自C公司的Mighty Amp试剂盒阳性检出率最高,达到95.83%;3种试剂盒检测下的阳性结果经RT-PCR检测,均为阳性。另外,检测24例阴性参比血清,结果显示均为阴性。3种试剂盒阳性检出率相比差异无统计学意义(P>0.05)。结论免核酸提取PCR试剂盒应用于乙型肝炎病毒的检测,方便快捷、特异性强、灵敏度高。Objective To select three kinds of PCR kits for the detection of hepatitis B virus. Methods 24 serum samples of hepatitis B patients and 24 negative serum samples were collected and detected by three kinds of PCR extraction kits. The sensitivity and specificity of the three kits were compared. The results were based on the fluorescence quantitative PCR results. Results The positive rate of MightyAmp kit purchased from C company was the highest, reaching 95.5%. The positive results of the three kits were all positive by RT-PCR, and 24 cases of negative reference serum were detected. The results showed that there was no significant difference in the positive detection rate of all kinds of test kits（P 〉 0.05）. Conclusion The PCR kit is convenient and rapid in the detection of hepatitis B virus. It is worth popularizing and applying in clinic because of its strong specificity and high sensitivity.
