摘 要:目的:观察肾复康对肾纤维化模型大鼠肾组织BMP-7表达的影响,探讨肾复康抗肾间质纤维化的作用机制。方法:40只SD大鼠随机分为假手术组、模型组、肾复康组和缬沙坦组,每组10只。采用单侧输尿管梗阻建立大鼠肾间质纤维化模型,分别给药治疗21d后处死大鼠,观察比较各组大鼠血Scr、BUN的变化,采用Masson染色观察肾组织纤维化程度,免疫组化法观察肾组织BMP-7的表达。结果:与假手术组比较,模型组血Scr、BUN水平明显升高(P<0.01),提示造模成功;肾复康组、缬沙坦组血Scr、BUN水平较模型组降低(P<0.01),2组间比较,差异亦有统计学意义(P<0.05)。肾复康组、缬沙坦组肾组织BMP-7表达较模型组增多(P<0.01),2治疗组间比较差异无统计学意义(P>0.05)。结论:肾复康能改善肾纤维化模型大鼠肾功能,减轻肾间质纤维化,其作用机制可能与调节肾组织BMP-7表达相关。Objective : To investigateeffect of Shenfukang on the expression of bone morphogenetic protein - 7 ( BMP - 7) in renal tissue in a rat model of renal fibrosis and the mechanism of action of anti - renal interstitial fibrosis effect of Shenfukang. Methods : A total of 40 Sprague - Dawley rats were randomly divided into sham - operation group, model group, Shenfukang group ,and valsartan group ,with 10 rats in each group. Unilateral ureteral obstruction was performed to establish a rat model of renal interstitial fibrosis. The rats were sacrificed after being treated with drugs for 21 days. The changes in serum creatinine (Scr) and blood urea nitrogen (BUN) were compared between groups, Masson staining was performed to observe the degree of renal fibrosis, and immnnohistochemistry was used to observe the expression of BMP - 7 in renal tissue. Results:Compared with the sham - operation group, the model group had significant increases in Scr and BUN (P 〈 0.01 ) , suggesting that the model was established successfully. Compared with the model group, the Shenfukang group and the valsartan group had significant reductions in Scr and BUN, and there were also significant differences between the two groups (P 〈 0. 05 ). Compared with the model group, the Shenfukang group and the valsartan group had a significant increase in the expression of BMP - 7 in renal tissue ( P 〈 0. 01 ) , and there was no significant difference between the two groups ( P 〉 0. 05 ). Conclusion : Shenfukang can improve renal function and alleviate renal interstitial fibrosis in a rat mod- el of renal fibrosis, possibly by regulating the expression of BMP - 7 in renal tissue.
