芬太尼的人体药物动力学及生物等效性研究

摘　　要：目的评价河南羚锐制药股份有限公司生产的芬太尼透皮贴剂与西安杨森制药有限公司进口比利时JANSSEN PHARMACEUTICA NV生产的芬太尼透皮贴剂(多瑞吉)人体生物利用度。方法以24名健康志愿者随机双周期交叉设计,采用HPLC-MS-MS法测定人血浆中芬太尼浓度。结果试验制剂与参比制剂的主要药动学参数Cmax分别为(468.7±227.4)和(508.6±261.6)pg·m L-1,Tmax分别为(35.1±19.0)和(38.2±21.1)h,AUC0~144h分别为(28132.6±12787.4)和(31609.3±16326.9)pg/(m L·h),AUC0→∞分别为(30251.2±14799.1)和(33063.0±17603.0)pg/(m L·h)。试验制剂对参比制剂的相对生物利用度F(以AUC0~144h作为评价依据)为94.0%±19.5%。结论芬太尼透皮贴剂试验制剂与参比制剂生物等效。OBJECTIVE To evaluate the bioavailability of transdermal fentanyl（ Durogesic） between Lingrui Pharmaceutical Co. Ltd（ Henan） and Xi＇an-Janssen Pharmaceutical Ltd imported from Belgia（ JANSSEN PHARMACEUTICA NV）. METHODS Crossover design,24 healthy volunteers in a randomized double-cycle,using HPLCMS-MS to determine the fentanyl concentration in human plasma. RESULTS The main pharmacokinetic parameters of the two transdermal patches of fentanyl were：Cmaxwere（468. 7 ± 227. 4） and（508. 6 ± 261. 6） pg·m L-1,Tmax were（35. 1 ± 19. 0） and（38. 2 ± 21. 1） h,AUC0 ~ 144 hwere（28132. 6 ± 12787. 4） and（31609. 3 ± 16326. 9） pg/（ h·m L）,AUC0→∞were（30251. 2 ± 14799. 1） and（33063. 0 ± 17603. 0） pg/（ h·m L）.The relative bioavailability of AUC0→144 was 94. 0% ± 19. 5%. CONCLUSION Two transdermal patches of fentanyl are bioequivalent.
